川崎病患儿外周血miR-145和心肌素的表达及其与冠状动脉损害的关系 被引量:5

Expressions of miR-145 and Myocardin in Peripheral Blood of Children with Kawasaki Disease and Their Relationships with Coronary Artery Lesion

摘要 目的研究微小核糖核酸-145(miR-145)和心肌素在川崎病患儿血清中的表达水平,分析二者与合并冠状动脉损害的关系。方法选取眉山市妇幼保健院和西南医科大学附属医院2015年5月—2018年10月收治的川崎病患儿72例为研究对象,按照是否出现冠状动脉损害分为无损害组(40例)和损害组(32例),另选取同期在眉山市妇幼保健院体检的健康志愿者40名为对照组。收集3组研究对象临床生化数据及血清炎症相关因子水平,采用酶联免疫吸附法检测血清miR-145和心肌素水平。结果冠状动脉损害组患儿血清C反应蛋白(CRP)、白细胞计数(WBC)、红细胞沉降率(ESR)、血小板计数(PLT)及白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)水平高于冠状动脉无损害组和对照组(P<0.05),且冠状动脉无损害组高于对照组(P<0.05)。冠状动脉损害组患儿血清miR-145及心肌素水平低于冠状动脉无损害组和对照组(P<0.05),且冠状动脉无损害组低于对照组(P<0.05)。miR-145在川崎病患儿血清中表达水平与心肌素水平呈正相关(P<0.01)。Logistic回归分析显示,低水平的血清miR-145、心肌素是川崎病患儿并发冠状动脉损害的危险因素(P<0.05)。结论miR-145、心肌素在川崎病合并冠状动脉损害患儿血清中低表达,低水平的血清miR-145、心肌素是川崎病患儿并发冠状动脉损害的危险因素。 Objective To study the expression of micro RNA-145(miR-145)and myocardin in serum of children with Kawasaki disease,and to analyze their relationships with coronary artery lesion.Methods Seventy-two children with Kawasaki disease from May 2015 to October 2018 were selected,and divided into non-coronary artery lesion group(40 cases)and coronary artery lesion group(32 cases),another 40 healthy volunteers in the same period were selected as control group.Clinical biochemical data and levels of serum inflammatory factors were collected,levels of serum miR-145 and myocardin were measured by ELISA.Results The levels of serum C-reactive protein(CRP),white blood cell count(WBC),erythrocyte sedimentation rate(ESR),platelet count(PLT),interleukin-6(IL-6),interleukin-8(IL-8),interleukin-10(IL-10),and tumor necrosis factor-α(TNF-α)of children in coronary artery lesion group were significantly higher than those in non-coronary artery lesion group and control group(P<0.05),data in non-coronary artery lesion group were significantly higher than those in control group(P<0.05).The levels of serum miR-145 and myocardin in coronary artery lesion group were significantly lower than those in non-coronary artery lesion group and control group(P<0.05),and the non-coronary artery lesion group was significantly lower than those in control group(P<0.05).The expression of miR-145 in serum of children with Kawasaki disease was positively correlated with myocardin(P<0.05).Conclusion The expressions of miR-145 and myocardin in serum of children with Kawasaki disease and coronary artery disease are low,decrease of serum miR-145 and myocardin is risk factors for coronary artery lesion in children with Kawasaki disease.
