摘要
目的探讨腹泻型肠易激综合征(IBS-D)患儿大肠黏膜组织中miR-143-5p水平与炎症反应及肠道微生物的关系。方法选取2017年1月-2019年1月收治的72例IBS-D患儿为研究组,同期的60例健康儿童为对照组。采用荧光定量PCR检测大肠黏膜组织中miR-143-5p水平,比较两组儿童大肠黏膜组织中miR-143-5p水平、肠道微生物数量、炎症因子水平,分析miR-143-5p水平与炎症反应及肠道微生物的关系。结果研究组患儿大肠黏膜组织中miR-143-5p水平高于对照组,双歧杆菌、乳酸杆菌、类杆菌数量低于对照组,肠杆菌数量高于对照组,血清肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-17(IL-17)水平高于对照组,血清白细胞介素-10(IL-10)水平低于对照组,差异均有统计学意义(P<0.05)。研究组患儿大肠黏膜组织中miR-143-5p水平与血清TNF-α、IL-1β、IL-17水平呈正相关,与血清IL-10水平呈负相关,与肠杆菌数量呈正相关,与双歧杆菌、乳酸杆菌、类杆菌数量呈负相关,与肠球菌无相关性。结论IBS-D患儿大肠黏膜组织中miR-143-5p水平升高,双歧杆菌、乳酸杆菌、类杆菌数量减少,肠杆菌数量增加,血清TNF-α、IL-1β、IL-17水平升高,血清IL-10水平降低,miR-143-5p表达与炎症反应及肠道微生物失调密切相关。
Objective To explore the relationships between miR-143-5p level and inflammatory response,intestinal microorganisms in colorectal mucosal tissue of children with diarrhea-predominant irritable bowel syndrome(IBS-D).Methods From January 2017 to January 2019,72 children with IBS-D treated in the hospital were selected as study group,and 60 healthy children were selected as control group.Fluorescent quantitative PCR was used to detect the level of miR-143-5p in colorectal mucosal tissue.The levels of miR-143-5p and inflammatory factors,the numbers of intestinal microorganisms in the two groups were compared.The relationships between miR-143-5p level and inflammatory response,intestinal microorganisms were analyzed.Results The level of miR-143-5p in colorectal mucosal tissue in study group was statistically significantly higher than that in control group,the numbers of Bifidobacterium,Lactobacillus,and Bacteroides in study group were statistically significantly lower than those in control group,the number of Enterobacterium in study group was statistically significantly higher than that in control group,the levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-17(IL-17)in study group were statistically significantly higher than those in control group,the level of interleukin-10(IL-10)in study group was statistically significantly lower than that in control group(P<0.05).The level of miR-143-5p in colorectal mucosal tissue in study group was positively correlated with serum TNF-α,IL-1β,and IL-17,the number of Enterobacterium;the level of miR-143-5p in colorectal mucosal tissue in study group was negatively correlated with serum IL-10,the numbers of Bifidobacterium,Lactobacillus,and Bacteroides,there was no correlation between miR-143-5p level and Enterococcus.Conclusion The level of miR-143-5p in colorectal mucosal tissue of children with IBS-D increases,the numbers of Bifidobacterium,Lactobacillus,and Bacteroides decrease,the number of Enterobacterium increases,the levels of serum TNF-α,IL-1β,and IL-17 increase,the level of serum IL-10 decreases,miR-143-5p expression is closely correlated with inflammatory reaction and intestinal microbial imbalance.
