HPLC测定不同形态西南手参中天麻素和对羟基苯甲醇的含量

Determination of the contents of gastrodin and 4-hydroxybenzyl alcohol in Gymnadeniae Rhizome of different morphology by HPLC

目的:测定不同形态西南手参中天麻素和对羟基苯甲醇的含量,研究西南手参形态及生长年限与这两种成分含量之间的关系。方法:色谱柱为HypersilGOLD C18色谱柱(250 mm×4.6 mm,5μm),以甲醇-0.1%磷酸水溶液(5∶95)为流动相,流速1.0 mL·min^-1,检测波长220 nm,柱温为30℃,进样量10μL。结果:天麻素和对羟基苯甲醇的进样量分别在0.00900~0.45000μg(r=0.9999)和0.00902~0.45100μg(r=1.0000)范围内与峰面积呈良好线性关系;天麻素和对羟基苯甲醇的平均加样回收率分别为96.72%和103.68%,RSD分别为2.08%和2.30%。三批样品中天麻素和对羟基苯甲醇总含量为0.4703~0.5500 mg·g^-1;随着指状分枝的增多及生长年限的增长,两批样品中天麻素和对羟基苯甲醇的总含量呈现逐渐升高的趋势,一批样品中天麻素和对羟基苯甲醇的总含量呈现先升后降的趋势。结论:西南手参中天麻素和对羟基苯甲醇的含量与其形态及生长年限有相关性。

Objective:To detect the contents of gastrodin and 4-hydroxybenzyl alcohol in Gymnadeniae Rhizome of different morphology and explore the relationship between morphology as well as growth year of Gymnadeniae Rhizome and the contents of the two components.Method:The sample were separated on Hypersil GOLD C 18 column(250 mm×4.6 mm,5μm),and the mobile phase was methanol-0.1%phosphoric acid(5:95).The flow rate was 1.0 mL·min^-1.The detection wavelength was set at 220 nm and the column temperature was maintained at 30℃.The sample size was 10μL.Result:The injection amounts of gastrodin and 4-hydroxybenzyl alcohol showed a good linear relationship with the peak area in the range of 0.00900~0.45000μg(r=0.9999)and 0.00902~0.45100μg(r=1.0000)respectively.The average recoveries of gastrodin and 4-hydroxybenzyl alcohol were 96.72%and 103.68%with the RSD of 2.08%and 2.30%.The total contents of gastrodin and 4-hydroxybenzyl alcohol in three batches of samples was 0.4703~0.5500 mg·g^-1.With the increase of the number of finger-like branches and the age of growth,the total contents of gastrodin and 4-hydroxybenzyl alcohol in two batches of samples showed a trend of gradual increase,while the total content in one batch of samples displayed a trend of rising at first and then reducing.Conclusion:The contents of gastrodin and 4-hydroxybenzyl alcohol in Gymnadeniae Rhizomaare correlated with its morphology and growing year.
