低剂量林丹暴露对大鼠睾丸生精细胞Bcl-2、CytC表达的影响 被引量:2

摘要 目的研究低剂量林丹暴露对SD大鼠睾丸生精细胞Bcl-2、CytC等蛋白表达的影响。方法选取健康的清洁级SD大鼠40只,随机分为5组,每组8只。参照大鼠林丹LD50(126 mg·kg-1),低、中、高剂量组按照每1 kg大鼠体质量分别给予3、6和13 mg的林丹,溶媒对照组给予10 mL玉米油,阳性对照组给予100μg雌二醇。以灌胃的方式连续染毒8周,测定大鼠睾丸附睾质量、系数及睾丸生精细胞Bcl-2、CytC的阳性表达情况。结果各实验组大鼠体质量增长量与溶媒对照组、阳性对照组比较差异均无统计学意义(P均>0.05);各实验组睾丸、附睾系数及睾丸质量与溶媒对照组、阳性对照组比较差异均无统计学意义(P均>0.05);各实验组附睾质量均高于阳性对照组(P均<0.05);高剂量组Bcl-2阳性表达低于溶媒对照组(P<0.05),低、中剂量组Bcl-2阳性表达高于阳性对照组(P<0.05);高剂量组CytC阳性表达高于溶媒对照组(P<0.05),低、中剂量组CytC阳性表达低于阳性对照组(P<0.05)。结论林丹可能通过下调睾丸组织中Bcl-2蛋白表达、上调CytC蛋白表达来调控生精细胞凋亡。 Objective To investigate the effects of low-dose lindane on the expression of Bcl-2 and CytC in spermatogenic cells of SD rats.Methods Forty healthy SD rats were randomly divided into 5 groups,8 in each group.According to the LD50(126 mg·kg-1)of lindane,3 mg,6 mg,and 13 mg of lindane were administered to the low,medium,and high dose groups,respectively,and 10mL of corn oil was administered to the solvent control group,and 100μg of estradiol was administered to the positive control group.The rats were continuously exposed to the drug for 8 weeks,and the testicular epididymis weight,coefficient and the positive expression rate of testicular spermatogenic cells Bcl-2 and CytC were determined.Results There was no difference in the body weight of each experimental group compared with the solvent control group and the positive control group(P all>0.05).There was no difference between the testis,epididymis coefficient and testicular weight of each experimental group compared with the solvent control group and the positive control group(P all>0.05).The weight of epididymis in each experimental group was higher than the positive control group(P all<0.05).The positive expression rate of Bcl-2 in the high dose group was lower than the solvent control group(P<0.05),and the positive expression rate of Bcl-2 in low and medium dose groups was higher than the positive control group(P<0.05).The positive expression rate of CytC in the high dose group was higher than the solvent control group(P<0.05),and the positive expression rate of CytC in the low and middle dose groups was lower than the positive control group(P<0.05).Conclusion Lindane may regulate the apoptosis of spermatogenic cells by down-regulating the expression of Bcl-2 protein and up-regulating the expression of CytC protein in the testis tissue.
