3种半边莲提取物体外抗猪繁殖与呼吸综合征病毒的作用研究 被引量:13

摘要 为研究半边莲水提取物、半边莲总黄酮、半边莲总生物碱体外抗猪繁殖与呼吸综合征病毒(PRRSV)的作用,本试验建立了Marc-145细胞模型,在研究其对Marc-145细胞最大安全质量浓度的基础上,结合细胞病变形态观察和MTT法,设立药物组、利巴韦林阳性对照组、病毒对照组和细胞对照组,对病毒进行阻断、抑制和直接杀灭3种作用方式的测定。结果显示,半边莲水提取物、半边莲总黄酮、半边莲总生物碱和利巴韦林的最大安全质量浓度分别为2.5、1.25、0.63和0.016 mg·mL^-1。在体外抗PRRSV作用的试验中,半边莲水提取物对病毒的阻断、抑制和直接灭活作用的最高抑制率分别为52.1%、60.2%、52.6%,极显著低于阳性对照利巴韦林(P<0.01),且各作用方式下均出现明显的细胞病变。半边莲总黄酮对病毒的阻断、抑制和直接灭活作用的最高抑制率分别为51.1%、85.0%、93.9%,其抑制和直接灭活作用较强,极显著高于阳性对照利巴韦林(P<0.01),且在这两种作用方式下细胞基本保持单层完好。半边莲总生物碱对病毒的阻断、抑制和直接灭活作用的最高抑制率分别为91.2%、57.8%、58.1%,其对PRRSV在抑制作用和直接灭活作用下的抑制率显著低于阳性对照利巴韦林(P<0.05),对PRRSV阻断作用极显著强于阳性对照利巴韦林(P<0.01),且在阻断作用下细胞轻微病变。综合3种作用方式,3种半边莲提取物均具有体外抗PRRSV的作用,其中,半边莲总黄酮抗PRRSV的效果最好,其次是半边莲总生物碱,半边莲水提取物抗PRRSV效果最弱。 In order to study the in vitro anti-porcine reproductive and respiratory syndrome virus(PRRSV)effect of the water extract,the total flavonoids and alkaloids of Lobelia Chinensis Lour,this experiment established the Marc-145 cell model.Based on the study of the maximum safe mass concentration of Marc-145 cells,and combined with cytopathic morphology observation and MTT method,the drug group,ribavirin positive control group,virus control group and cell control group were set up to determine the three action modes of blocking,inhibition and direct inactivation of the virus,respectively.The results showed that the maximum safe concentrations of the water extract,the total flavonoids,the total alkaloids of Lobelia Chinensis Lour and ribavirin were 2.5,1.25,0.63 and 0.016 mg·mL^-1,respectively.In the experiment of anti-PRRSV effect in vitro,the highest inhibition rates of the blocking,inhibition and direct inactivation of the water extract of Lobelia Chinensis Lour were 52.1%,60.2%,and 52.6%,respectively,which were very significantly lower than those of the positive control ribavirin(P<0.01),and there appeared obvious cytopathic effect under the three modes of action.The highest inhibition rates of the blocking,inhibition and direct inactivation of the total flavonoids of Lobelia Chinensis Lour were 51.1%,85.0%,and 93.9%,respectively.The inhibition and direct inactivation were stronger,very significantly higher than those of the positive control ribavirin(P<0.01).In these two modes of action,the cells remained essentially intact.The highest inhibition rates of blocking,inhibition and direct inactivation of the total alkaloids of Lobelia Chinensis Lour were 91.2%,57.8%,and 58.1%,respectively,the inhibition rate of PRRSV under the inhibition and direct inactivation was significantly lower than that of the positive control ribavirin(P<0.05),and the blocking effect on PRRSV was significantly stronger than that of the positive control ribavirin(P<0.01).The cells were slightly pathological under the action of blocking.In combination with the three modes of action,the three kinds of extracts of Lobelia Chinensis Lour all had in vitro anti-PRRSV effects.Among them,the total flavonoid of Lobelia Chinensis Lour had the best anti-PRRSV effect,followed by the total alkaloids,and the water extract had the weakest anti-PRRSV effect.
