小鼠海马神经元急性振动分离及电生理活性鉴定

摘要 目的:建立一种适用于膜片钳实验的小鼠特定脑区神经元的分离方法。方法:采用机械振动的方法分离小鼠海马CA1区锥体细胞,利用穿孔膜片钳记录细胞自发和诱发的动作电位,观察电压和受体依赖性离子通道电流,评估分离方法效果。结果:分离得到的海马CA1区锥体细胞保留了基本形态特征,具有正常的电生理学特性。结论:急性振动分离法是一种适用于分离小鼠脑区神经元的方法,分离的神经元适用于膜片钳实验。 Objective:To establish a method for isolating neurons from specific brain regions of mice suitable for patch clamp experiment.Methods:Mechanical vibration method was used to isolate the pyramidal cells in CA1 area of mouse hippocampus.The spontaneous and induced action potentials were recorded by using perforated patch clamp.The voltage and receptor dependent ion channel current were observed to evaluate the effect of the separation method.Results:The pyramidal cells in CA1 area of hippocampus preserved the basic morphological characteristics and had normal electrophysiological characteristics.Conclusion:The method of acute vibration separation is suitable for isolating neurons in the brain of mice,and the isolated neurons are suitable for patch clamp experiment.
