摘要
目的:研究太岁水提物(tai sui aqueous extracts,TSAE)对衰老皮肤透明质酸(hyaluronic acid,HA)和表皮角蛋白K19的影响.方法:用D-半乳糖构建衰老大鼠模型,同时分别给予不同剂量的(0、50、100和200 mg/kg)的TSAE处理,利用HE染色法观察大鼠皮肤组织结构的变化,用酶联免疫吸附(ELISA)法检测皮肤组织中透明质酸质量浓度,定量PCR和Western blot检测皮肤组织中透明质酸结合蛋白1(hyaluronan binding protein1,HABP1)、表皮角蛋白K19 mRNA水平和蛋白含量.结果:与对照组比较,模型组皮肤出现明显的病理性改变,HA质量浓度、HABP1、表皮角蛋白K19表达水平显著下降.与模型组比较,TSAE干预后,大鼠皮肤病理改变明显减轻,HA质量浓度、HABP1、表皮角蛋白K19表达水平明显上调.结论:TSAE对衰老皮肤有保护作用,机制可能与其干预HA含量及表皮角蛋白K19的表达有关.
Objective The aims of this study was to investigate the effects of the TaiSui aqueous extracts(TSAE)on the hyaluronic acid(HA)and epidermal keratin 19 in the aging skin.Methods Aging rat models were made by D-galactose,and then treated continuously with different doses(0,50,100,200 mg/kg)of TSAE.The changes of skin histological structure were observed under microscope by HE staining.The content of HA in skin tissues was detected by ELISA method.Quantitative PCR and Western blotting were used to detect mRNA and protein levels of HABP1 and K19 in the skin tissues.Results In comparison with control group,there were obvious pathological changes in the skin histological structure of the rats model group,and the content of HA,expressions of HABP1 and K19 were statistically down-regulated.Compared to model group,administration of TSAE could obviously alleviate skin pathological changes.The levels of HABP1 and K19 were markedly increased.Conclusions TSAE had protective effect on skin aging,and the mechanism may be related to interfering with the content of HA and expression levels of K19 in the skin.
