摘要 目的探讨细胞分裂周期相关蛋白(CDCA2)基因在原发性肝细胞癌中的表达情况及其临床意义。方法通过下载癌症基因组(TCGA)数据库相关数据,分析CDCA2基因与原发性肝细胞癌患者临床病理特征以及生存预后的关系。结果总体363例原发性肝细胞癌样本的癌和癌旁组织中的CDCA2的表达水平分别为(5.3106±1.6239)和(2.2912±1.224),差异有统计学意义(t=75.04,P<0.000)。50例配对的原发性肝细胞癌和癌旁组织中的CDCA2表达水平分别为(5.0955±1.5494)和(2.2912±1.224),差异有统计学意义(t=12.921,P<0.001)。ROC曲线分析提示CDCA2基因可作为一个灵敏度和特异度较高的原发性肝细胞癌诊断指标(敏感性84.0,特异性90.0,AUC 0.929)。CDCA2基因表达与原发性肝细胞癌患者的年龄、AFP值、病理分级、T分期、M分期、临床分期均有关(P<0.05),但与患者性别、肝功能分级、N分期无关(P>0.05)。生存分析发现总体363例样本中CDCA2低表达者和高表达的总体生存时间(OS)分别为(938.98±777.51)和(721.15±679.54)d,差异具有统计学意义(Log-rank=32.58,P<0.001)。无复发生存期(RFS)分别为(698.25±658.37)和(522.55±570.88)d,差异有统计学意义(Log-rank=32.58,P<0.001)。结论CDCA2在原发性肝细胞癌中高表达,且临床特征密切相关,其高表达对判断原发性肝细胞癌的生存预后及其诊断具有参考价值。 Objective To investigate the expression and clinical significance of CDCA2 gene in primary hepatocellular carcinoma.Methods The relationship between CDCA2 gene and clinicopathological features and survival prognosis in patients with primary hepatocellular carcinoma was analyzed by downloading data from the Cancer Genome Atlas(TCGA)database.Results The expression levels of CDCA2 in cancer and paracancerous tissues of 363 primary hepatocellular carcinoma samples were(5.3106±1.6239)and(2.2912±1.224),respectively,and there were statistical significantly differences(t=75.04,P<0.000).The expression levels of CDCA2 in 50 matched primary hepatocellular carcinomas and adjacent tissues were(5.0955±1.5494)and(2.2912±1.224),respectively,and there were statistical significantly differences(t=12.921,P<0.001).ROC curve analysis indicated that CDCA2 gene can be used as a diagnostic index for primary hepatocellular carcinoma with high sensitivity and specificity(sensitivity was 84.0,specificity was 90.0,AUC=0.929).CDCA2 gene expression and age,AFP,pathological grade(P=0.000),T stage,M stage,clinical stage were all related,there were statistical significantly differences,but it was not related to gender,liver function grading and N staging,and there were no statistical significantly differences(P>0.05).Survival analysis found that the OS of CDCA2 low expression group and high expression group were(938.98±777.51)and(721.15±679.54)days,respectively,and there were statistical significantly differences(Log-rank=32.58,P<0.001).The RFS was(698.25±658.37)and(522.55±570.88)days,respectively,and there were also statistical significantly differences(Log-rank=32.58,P<0.001).Conclusion CDCA2 is highly expressed in primary hepatocellular carcinoma,and its clinical features are closely related.Its high expression has reference value for judging the survival prognosis and diagnosis of primary hepatocellular carcinoma.
