CPS1与CPS1-IT1在肝内胆管癌患者中的表达及与病理的相关性研究

Expression of CPS1 and CPS1-IT1 in patients with intrahepatic cholangiocarcinoma and their correlation with pathology

目的:探讨氨甲酰磷酸合成酶(CPS1)与氨甲酰磷酸合成酶1-内含子转录本1(CPS1-IT1)在肝内胆管癌患者中的表达及与病理的相关性。方法:选择2014年4月至2019年4月肝内胆管癌(ICC)患者36例作为研究对象,所有患者均行手术治疗,术中分别取患者的癌旁肝组织及癌组织,分为对照组和观察组。记录并统计患者性别、年龄、肿瘤分期、淋巴结转移、分化类型等病理资料,测定不同组织中CPS1、CPS1-IT1与CPS1+CPS1-IT1表达水平,分析不同病理状态下CPS1与CPS1-IT1的表达量,并进行多因素Logistic分析;采用SPSS Pearson相关性分析软件对ICC患者的CPS1与CPS1-IT1表达水平与病理情况进行相关性分析。结果:CPS1、CPS1-IT1与CPS1+CPS1-IT1在ICC癌组织和癌旁肝组织的相对表达量均有显著性差异(P<0.05);观察组患者ICC组织中CPS1与CPS1-IT1的表达水平与肿瘤分期、分化类型及TNM分期具有统计学意义(P<0.05);SPSS Pearson相关性分析结果表明,CPS1基因及CPS1-IT1的表达水平ICC患者肿瘤直径、淋巴转移、T分期呈正相关性(P<0.05),与分化类型呈负相关性(P<0.05)。结论:CPS1与CPS1-IT1在ICC组织内共同高表达,且与肿瘤分期、淋巴结转移、分化类型等存在相关性,CPS1与CPS1-IT1表达水平测定能反映疾病严重程度,指导临床治疗,并可能成为ICC临床诊断相对特异的肿瘤标志物。

Objective:to investigate the expression of CPS1 and CPS1-IT1 in patients with intrahepatic cholangiocarcinoma and its correlation with pathology.Methods:36 patients with intrahepatic cholangiocarcinoma(ICC)from April 2014 to April 2019 were selected as the subjects.All patientswere prepared for surgical treatment,and the para-carcinoma liver tissues and carcinoma tissues of the patients were collected intraoperatively and divided into control group and observation group.The expression levels of CPS1,CPS1-IT1 and CPS1+CPS1-IT1 in different tissues were measured.The pathological data of the observation group were consulted,and the gender,age,tumor stage,lymph node metastasis and differentiation type of the patients were counted.The expressions of CPS1 and CPS1-IT1 in different pathological conditions were analyzed,and multivariate Logistic analysis was completed.The correlation between ICC expression levels of CPS1 and CPS1-IT1 and pathological conditions was analyzed by SPSS Pearson correlation analysis software.Result:the relative expression levels of CPS1,CPS1-IT1 and CPS1+CPS1-IT1 in ICC and para-carcinoma tissues were significantly different(P<0.05).The expression levels of CPS1 and CPS1-IT1 in the ICC of the observation group were statistically significant with respect to tumor stage,differentiation type and TNM stage(P<0.05).The results of SPSS Pearson correlation analysis showed that the expression levels of CPS1 gene and CPS1-IT1 were positively correlated with tumor diameter,lymphatic metastasis and T stage in patients with intrahepatic cholangiocarcinoma(P<0.05),and negatively correlated with the differentiation type(P<0.05).Conclusion:CPS1 and CPS1-IT1 are both highly expressed in ICC cancer tissues and are correlated with tumor stage,lymph node metastasis,and differentiation type,etc.,so the enhancement of CPS1 and CPS1-IT1 expression level can reflect the severity of disease,guide clinical treatment,and may become relatively specific tumor markers for ICC clinical diagnosis.
