摘要
为建立测定桔梗茎、叶中总皂苷含量的方法,通过比较香草醛-冰醋酸-浓硫酸比色法、香草醛-冰醋酸-高氯酸比色法、香草醛-乙醇-硫酸比色法的紫外可见光谱图,确定香草醛-冰醋酸-浓硫酸比色法更适合测定桔梗茎、叶中总皂苷含量,并对该方法的显色条件进行了优化,同时对其进行方法学验证。试验结果表明,该方法操作简便快速,专属性好,灵敏度高,重复性好,稳定性高,适用于桔梗茎、叶中总皂苷含量的测定。
A spectrophotometric method for the determination of total saponins in stem and leaf of Platycodon grandiflorum was established by comparing vanillin-aceticacid-H2S04 staining method,vanillin-aceticacid-HC1O4 staining method,vanillin-ethanol-H2S04 staining method to determine the content of total saponins in stem and leaf of Platycodon grandiflorum.It was found that the colorimetric method of vanillin-aceticacid-H2SO4 staining method was more suitable for the determination of total saponins in the stems and leaves of Platycodon grandiflorum,and the color conditions of the method were optimized.Meanwhile,its methodology is verified.The results show this method was simple,rapid,specific,sensitive,reproducible and stable.It was suitable for the determination of total saponins in stem and leaf of Platycodon grandiflorum.
