摘要
目的探讨RhoA/ROCK1在多发性骨髓瘤中的表达情况。方法选取该院2011年1月-2019年8月收治的30例MM患者作为观察组,依据骨质破坏程度分为早期破坏组11例和晚期破坏组19例,选取同期该院原发免疫性血小板减少症(ITP)患者20例作为对照组。结果观察组患者Rho A、ROCK1表达(3.87±0.97)ng/mL、(4.09±0.46)ng/mL和mRNA相对表达量(1.78±0.52)ng/mL、(1.33±0.33)ng/mL高于对照组(1.80±0.56)、(3.37±0.21)ng/mL和(1.01±0.07)、(1.01±0.09)ng/mL,差异有统计学意义(t=8.62、6.54、6.56、4.22,P<0.05)。早期破坏组(2.62±0.52)、(3.91±0.66)ng/mL,晚期破坏组(4.16±1.81)、(4.17±0.98)ng/mL。患者Rho A、ROCK1表达和mRNA相对表达量高于对照组,晚期破坏组Rho A、ROCK1表达和mRNA相对表达量高于早期破坏组,Rho A蛋白表达水平与Hb呈现负相关(r=-0.673,P=0.000),与αCa、β2-mg呈现正相关(r=0.464、P=0.002,r=0.339、P=0.019),ROCK1蛋白表达水平与Hb呈现负相关(r=-0.583,P=0.000),与Ca、αCa、β2-mg呈现正相关(r=0.356、0.420、0.341,P=0.021、0.011、0.030)。结论RhoA/ROCK1在多发性骨髓瘤中处于高表达水平,其和骨质破坏程度呈现明显正相关。
Objective To investigate the expression of RhoA/ROCK1 in multiple myeloma.Methods Thirty patients with MM treated in our hospital from January 2011 to August 2019 were selected as the observation group.According to the degree of bone destruction,they were divided into 11 early damage groups and 19 late damage groups.The primary immunity of our hospital during the same period was selected.20 patients with thrombocytopenia(ITP)served as the control group.Results The expressions of Rho A and ROCK1 in the observation group were(3.87±0.97)ng/mL,(4.09±0.46)ng/mL,and the relative expression levels of mRNA(1.78±0.52)ng/mL,(1.33±0.33)ng/mL were higher than control group(1.80±0.56)ng/mL,(3.37±0.21)ng/mL and(1.01±0.07)ng/mL,(1.01±0.09)ng/mL,the difference was statistically significant(t=8.62,6.54,6.56,4.22,,P<0.05),early destruction group(2.62±0.52)ng/mL(3.91±0.66)ng/mL,late destruction group(4.16±1.81)、(4.17±0.98)ng/mL had higher expressions of Rho A,ROCK1,and relative mRNA expression than those in the control group,the expression of Rho A,ROCK1 and the relative expression of mRNA in the late damage group were higher than those in the early damage group,Rho A protein expression level was negatively correlated with Hb(r=-0.673,P=0.000),showing positive correlation withαCa andβ2-mg(r=0.464、P=0.002,r=0.339、P=0.019),ROCK1 protein expression level showed negative correlation with Hb(r=-0.583,P=0.000),showing positive correlation with Ca,αCa,β2-mg(r=0.356、0.420、0.341,P=0.021、0.011、0.030).Conclusion RhoA/ROCK1 is highly expressed in multiple myeloma,and it is significantly positively correlated with the degree of bone destruction.
