摘要
LRH-1(nuclear receptor subfamily 5, group A, member 2),是核受体家族的成员之一,作为转录共激活子调控相关基因的表达,对类固醇激素的分泌及细胞代谢有重要影响。为探究LRH-1对牛卵巢颗粒细胞类固醇激素分泌的影响,以体外培养的牛卵巢颗粒细胞为试验材料,使用不同浓度(20μM、50μM、100μM、150μM、200μM)LRH-1的激活剂DLPC激活LRH-1的表达,通过免疫荧光、免疫组织化学和RT-qPCR方法,检测LRH-1在牛卵巢组织中的表达定位及其对牛卵巢颗粒细胞类固醇合成相关基因表达水平的影响。试验结果表明,LRH-1在牛卵巢颗粒细胞中高度表达,50μM DLPC可显著提高STAR、LRH-1、CYP17基因的表达,20μM DLPC可显著抑制CYP11基因的表达。研究结果揭示了LRH-1表达的上调可以显著提高类固醇激素合成相关基因的表达。
LRH-1(nuclear receptor subfamily 5,group A,member 2)is a member of the nuclear receptor family.It acts as a transcriptional coactivator to regulate the expression of related genes,and has an important influence on the secretion of steroid hormones and cellular metabolism.To investigate the effect of LRH-1 on the secretion of steroid hormones in bovine ovarian granulosa cells,this experiment used bovine ovarian granulosa cells cultured in vitro as experimental materials,using different concentrations(20μM,50μM,100μM,150μM,200μM)of LRH-1 activators DLPC to activate the expression of LRH-1,and the expression of LRH-1 in bovine ovarian tissue and its effect on the expression level of steroidogenic genes related to bovine ovarian granule cells were detected and studied by immunofluorescence,immunohistochemistry and RT-qPCR.The results showed that LRH-1 was highly expressed in bovine ovarian granulosa cells.50μM DLPC significantly increased the expression of STAR,LRH-1 and CYP17 genes,and 20μM DLPC significantly inhibited the expression of CYP11 gene.These findings revealed that up-regulation of LRH-1 expression can significantly increase the expression of steroidogenic synthesis-related genes.
