美罗培南对胞内不同c-di-GMP浓度表型铜绿假单胞菌生物被膜形成的影响 被引量:5

Effect of meropenem on biofilm formation of phenotype pseudomonas aeruginosa with different c-di-GMP concentrations in cells

摘要 目的:探究美罗培南对胞内不同环二鸟苷酸(c-di-GMP)浓度表型铜绿假单胞菌(PA)生物被膜形成的影响。方法:两倍稀释法测定美罗培南对各菌株的抑菌浓度(MIC);设阴性对照组和3个美罗培南干预组(1/2MIC、1/4MIC、1/8MIC)。用1/2MIC、1/4MIC、1/8MIC的美罗培南分别干预野生株PAO1、PA胞内c-di-GMP高表达株(PAO1ΔwspF)、PA胞内c-di-GMP低表达株(PAO1/Plac-yhjH),观察美罗培南对其运动能力的影响,用结晶紫染色法测定美罗培南对PAO1、PAO1ΔwspF、PAO1/Plac-yhjH菌株的生物被膜形成以及对生物被膜清除能力的影响。结果:在美罗培南的干预下,PAO1、PAO1ΔwspF、PAO1/Plac-yhjH的运动区域直径均比阴性对照组小(P<0.05),所形成的的生物被膜也均比阴性对照组少(P<0. 05),而破坏的生物被膜均比阴性对照组多(P<0. 05)。结论:美罗培南能抑制PAO1、PAO1ΔwspF、PAO1/Plac-yhjH的运动能力以及生物被膜形成,美罗培南能破坏PAO1、PAO1ΔwspF、PAO1/Plac-yhjH形成的生物被膜,美罗培南对PAO1、PAO1ΔwspF、PAO1/Plac-yhjH的抑制效果不同。 Objective:To investigate the effect of meropenem on the biofilm formation of phenotype pseudomonas aeruginosa(PA)with different concentrations of intracellular c-di-GMP.Methods:Two-fold dilution method was used to determine the minimum inhibitory concentration(MIC)of meropenem on each strain;a negative control group and three meropenem intervention groups(1/2 MIC,1/4 MIC,1/8 MIC)were set up.1/2 MIC,1/4 MIC,and 1/8 MIC meropenem were used to intervene in the wild strain PAO1,PA intracellular c-di-GMP high expression strain(PAO1Δwsp F),and PA intracellular c-di-GMP low expression strain(PAO1/Plac-yhj H)to observe the effect of meropenem on their exercise ability.The effect of meropenem on the biofilm formation of PAO1,PAO1Δwsp F,PAO1/Plac-yhj H strains and its effect of biofilm removal were detected by crystal violet staining.Results:Under the intervention of meropenem,the diameters of the exercise areas of PAO1,PAO1Δwsp F,and PAO1/Plac-yhj H were smaller than those of the negative control group(P<0.05),less biofilms were formed than in the negative control group(P<0.05),and more biofilms were destroyed than in the negative control group(P<0.05).Conclusion:Meropenem can inhibit the exercise ability and biofilm formation of of PAO1,PAO1Δwsp F,and PAO1/Plac-yhj H.Meropenem can destroy the biofilm formed by of PAO1,PAO1Δwsp F,and PAO1/Plac-yhj H.The inhibitory effects of meropenem on of PAO1,PAO1Δwsp F,and PAO1/Plac-yhj H are different.
