摘要
目的:探讨黄芪注射液后处理对兔心肌缺血再灌注损伤(MIRI)的保护作用及其机制。方法:40只雄性成年新西兰兔随机分为4组:假手术组(Sham组)、缺血再灌注组(IR组)、缺血后处理组(IPO组)和黄芪注射液后处理组(AS组),每组10只。在再灌注末抽取颈内静脉血测定血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、肌钙蛋白(cTnI)、丙二醛(MDA)和超氧化物歧化酶(SOD)的含量;留取心肌组织,采用高效液相色谱法测定心肌三磷酸腺苷(ATP)、二磷酸腺苷(ADP)和环酸腺苷(AMP)含量;检测心肌梗死面积。结果:与Sham组比较,IR组、IPO组和AS组血浆MDA、CK、LDH、cTnI和心肌组织ADP含量显著增高(P<0.05),SOD活性和心肌组织ATP含量显著降低(P<0.05);与IR组比较,IPO组和AS组血浆MDA、CK、LDH、cTnI和心肌组织ADP含量以及梗死心肌质量和梗死面积显著降低(P<0.05),SOD活性和心肌组织ATP含量显著增高(P<0.05);IPO组和AS组比较,差异无统计学意义(P>0.05)。结论:本实验条件下,黄芪注射液后处理可减轻兔MIRI,具有类似缺血后处理的心肌保护作用,其机制可能是通过减少氧自由基的生成,从而增强心肌抗氧化能力,改善心肌功能代谢。
Objective:To explore the protective effect and its mechanism of post-treatment of Astragalus injection on myocardial ischemia-reperfusion injury(MIRI)in rabbits.Methods:Forty male adult New Zealand rabbits were randomly divided into 4 groups:sham operation group(Sham group),ischemia-reperfusion group(IR group),ischemic post-treatment group(IPO group),and Astragalus injection post-treatment group(AS Group),with 10 cases per group.At the end of reperfusion,internal jugular venous blood was drawn to determine the content of serum creatine kinase(CK),lactate dehydrogenase(LDH),troponin(cTnI),malondialdehyde(MDA),and superoxide dismutase(SOD).Collected the myocardial tissue,the content of myocardial adenosine triphosphate(ATP),adenosine diphosphate(ADP),and cyclic acid adenosine(AMP)were detected using high-performance liquid chromatography.And the myocardial infarction area was detected.Results:Compared with Sham group,plasma MDA,CK,LDH,c TnI,and myocardial tissue ADP content were significantly increased(P<0.05),while SOD activity and myocardial tissue ATP content were significantly reduced(P<0.05)in IR group,IPO group,and AS group.Compared with IR group,the plasma MDA,CK,LDH,cTnI,and myocardial tissue ADP content,infarcted myocardial mass and infarct area were significantly reduced(P<0.05),while SOD activity and myocardial tissue ATP content were significantly increased(P<0.05)in the IPO group and the AS group.There was no statistically significant difference between the IPO group and the AS group(P>0.05).Conclusion:Under the conditions of this experiment,post-treatment of Astragalus injection can reduce MIRI in rabbits and has a myocardial protective effect similar to ischemic post-treatment.The mechanism may be reducing the generation of oxygen free radicals and thereby enhancing myocardial antioxidant capacity and improving myocardial function metabolism.
