摘要
目的:观察不同剂量维生素E对大鼠薄型子宫内膜的修复作用,初步探讨其可能的作用机制。方法:将50只SPF级健康性成熟雌性SD大鼠按随机数字表法分为正常组、模型组、低剂量组(15 mg/kg)、中剂量组(30 mg/kg)、高剂量组(60 mg/kg),每组10只。于4个动情期后的动情期取材,采用苏木精-伊红(HE)染色法观察各组大鼠子宫内膜厚度及腺体数目,免疫荧光法检测子宫内膜CD31的表达,酶联免疫吸附测定(ELISA)法检测血管内皮生长因子(VEGF)、白介素-10(IL-10)、雌激素、孕酮、白血病抑制因子(LIF)的表达。结果:与正常组相比,模型组大鼠子宫内膜厚度、腺体数目和VEGF、LIF、CD31的表达量均明显减少(P<0.01),高剂量组大鼠子宫内膜厚度、腺体数目和VEGF、CD31的表达量均明显增加(P<0.01);与模型组相比,中剂量组和高剂量组大鼠子宫内膜厚度、腺体数目和VEGF、IL-10、LIF、CD31的表达量均明显增加(P<0.01),雌激素、孕酮表达水平随维生素E剂量增加而增加(P<0.01)。结论:维生素E可能通过上调大鼠子宫内膜中CD31、VEGF的表达及调节卵巢雌孕激素的分泌从而改善子宫内膜厚度,并且上调IL-10的表达从而抑制创伤的炎症反应,以及提高子宫内膜容受性相关因子LIF的水平,进而改善子宫内膜容受性,发挥修复作用。
Objective:To observe the repair effect of different doses of vitamin E on the thin endometrium of rats,and to explore its possible mechanism.Methods:Fifty SPF healthy and mature female SD rats were divided into the normal group,the model group,the low-dose group(15 mg/kg),the medium-dose group(30 mg/kg),and the high-dose group(60 mg/kg)using a random number table,with 10 cases in each group.The tissue materials were collected at the estrus period after 4 estrus periods.Hematoxylin-eosin(HE)staining was used to observe the endometrial thickness and number of glands in each group of rats.Immunofluorescence method was used to detect the expression of endometrium CD31.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of vascular endothelial growth factor(VEGF),interleukin-10(IL-10),estrogen,progesterone,and leukemia inhibitory factor(LIF).Results:Compared with the normal group,the endometrium thickness,number of glands,and the expression of VEGF,LIF,and CD31 were significantly reduced in the model group(P<0.01);the endometrial thickness,gland number,and expression of VEGF and CD31 were significantly increased in the high-dose group(P<0.01).Compared with the model group,the endometrial thickness,number of glands,and expression of VEGF,IL-10,LIF and CD31 were significantlyincreased(P<0.01),and the expression levels of estrogen and progesterone increased with the increase of vitamin E dose in the medium-dose and high-dose groups(P<0.01).Conclusion:Vitamin E may improve the endometrial thickness by up-regulating the expression of CD31 and VEGF in the endometrium of rats and thus regulating the secretion of ovarian estrogen and progesterone,inhibit the inflammatory reaction of wounds by up-regulating the expression of IL-10 andplay a repairing role by increasing the level of endometrial receptivity-related factor LIF.
