SIRT1基因与肺癌免疫浸润水平及临床预后分析 被引量:4

摘要 目的:通过生物信息学分析沉默信息调节因子2相关酶1(SIRT1)基因在肺癌与正常组织的差异表达,并分析其与肺癌预后和免疫浸润水平的关系。方法:通过生物信息数据库获取SIRT1基因在肺癌中的表达情况,并分析SIRT1基因与肺癌肿瘤免疫浸润水平及预后的影响。结果:与正常组织相比,SIRT1基因在肺癌中表达下降(P<0.05)。SIRT1基因的表达与肺癌患者中B细胞、CD8+T细胞、CD4+T细胞、巨噬细胞、嗜中性粒细胞、树突状细胞的免疫浸润水平呈正相关关系(P<0.05)。SIRT1基因的表达还与肺癌中多种免疫标志物关系密切。此外,SIRT1基因的低表达与肺癌的整体生存率(OS)、首次渐进生存期(FP)及再次进展生存期(PPS)相关(OS HR=0.7,P=0.000;FP HR=0.75,P=0.004;PPS HR=0.73,P=0.017)。结论:SIRT1基因在肺癌中表达下调,从而影响肺癌肿瘤免疫浸润细胞水平,导致免疫微环境失衡,进而造成不良的临床预后。这些发现表明,SIRT1可用作确定肺癌的预后和免疫浸润的预后生物标志物。 Objective:To analyze the differential expression of silencing information regulator 2 related enzyme1(SIRT1)gene in lung cancer and normal tissues by bioinformatics,and to analyze its relationship with the prognosis of lung cancer and the level of immune infiltration.Methods:The expression of SIRT1 gene in different types of cancer were obtained through the biological information database.Analyzedof the influence of SIRT1 gene on the level of the immune infiltration,clinicopathological characteristics,and prognosis of the tumor.Results:Compared with normal tissues,expression ofSIRT1 gene was decreased in lung cancer(P<0.05).The low expression of SIRT1 gene was positively correlated with the immune infiltration level of B cells,CD8+T cells,CD4+T cells,macrophages,neutrophils,and dendritic cells in patients with lung cancer.The mutation of the copy number of SIRT1 gene was closely related to the level of tumor immune infiltrating cells(P<0.05).The expression of SIRT1 gene was also closely related to multiple immune markers in lung cancer.In addition,the low expression of SIRT1 gene was associated with the overall survival rate(OS),first progressive survival(FP),and progressive progression survival(PPS)of patients with lung cancer(OS HR=0.7,P=3.3 e-08;FP HR=0.75,P=0.0039;PPS HR=0.73,P=0.017).Conclusion:The expression of SIRT1 gene is down-regulated in lung cancer,which affects the level of immune infiltrating cells of lung cancer tumors,resulting in an imbalance of the immune microenvironment,which in turn leads to poor clinical prognosis.These findings indicate that SIRT1 can be used as a biomarker to in the prognosis and immune infiltrationof lung cancer.
