摘要
目的:分析MDC1对卵巢癌细胞迁移、侵袭与凋亡的影响,及其可能的作用机制。方法:采集我院2015年8月~2016年10月收治的80例卵巢癌患者临床标本及细胞系人卵巢HO8910PM标本,采用免疫组化分析MDC1与卵巢癌进展预后关系,MTT试验、细胞划痕法与MDC1小干扰RNA(siRNA)序列研究MDC1对卵巢癌细胞增殖侵袭的影响,流式细胞仪分析细胞周期与凋亡情况。结果:免疫组化检测结果显示,MDC1主要表达在细胞核中;生存曲线分析表明:与MDC1低表达组比较,MDC1高表达组的预后显著较差(P<0.05)。Transwell实验及划痕实验均表明:对照组相比,低表达MDC1的细胞迁移和侵袭能力降低(P<0.05)。流式细胞术检测结果表明MDC1可促进卵巢癌细胞的增殖与细胞周期的进展(P<0.05)。结论:MDC1的表达水平可能与卵巢癌预后相关,其高表达提示预后差,同时MDC1具有促进卵巢癌迁移、侵袭的作用,并促进卵巢癌细胞增殖与细胞周期的进展。
Objective:To analyze the effect of MDC1 on migration,invasion and apoptosis of ovarian cancer cells,and its possible mechanism of action.Methods:The clinical specimens and cell ovary HO8910PM specimens from 80 patients with ovarian cancer admitted in our hospital from August 2015 to October 2016 were evaluated.Immunohistochemistry was used to analyze the relationship between MDC1 and prognosis of ovarian cancer,MTT assay,cell scratch method and MDC1 small interfering RNA(siRNA)sequence were used to study the effect of MDC1 on proliferation and invasion of ovarian cancer cells,and flow cytometry was used to analyze cell cycle and apoptosis.Results:The results of immunohistochemistry showed that MDC1 was mainly expressed in the nucleus.Survival curve analysis showed that the prognosis of MDC1 high expression group was significantly worse than that of MDC1 low expression group(P<0.05).Transwell experiments and scratch experiments showed that the migration and invasion ability of cells with low expression of MDC1 was lower than that of the control group(P<0.05).Flow cytometry results showed that MDC1 could promote the proliferation and cell cycle progression of ovarian cancer cells(P<0.05).Conclusion:The expression level of MDC1 may be related to the prognosis of ovarian cancer,and its high expression suggests poor prognosis,while MDC1 promotes the migration and invasion of ovarian cancer and promotes the proliferation of ovarian cancer cells and the progression of the cell cycle.
