巨噬细胞来源的TGF-β对小鼠骨髓成纤维细胞增殖及Ⅰ型胶原表达的影响

摘要 目的:研究巨噬细胞表达分泌的TGF-β对小鼠原代骨髓成纤维细胞增殖的影响,揭示巨噬细胞在骨髓纤维化疾病进展中的作用。方法:分离培养小鼠骨髓来源巨噬细胞(BMMs)和骨髓成纤维细胞,利用LPS刺激巨噬细胞活化并收集巨噬细胞条件培养基;ELISA检验细胞TGF-β和Ⅰ型胶原表达;MTT法检测TGF-β和巨噬细胞条件培养基对骨髓成纤维细胞增殖的影响。结果:LPS刺激巨噬细胞活化并表达TGF-β;TGF-β及巨噬细胞条件培养基均能促进骨髓成纤维细胞增殖及Ⅰ型胶原表达;使用抗体中和培养基中的TGF-β可以抑制骨髓成纤维细胞的增殖。结论:表达分泌的TGF-β可以促进骨髓成纤维细胞增殖和Ⅰ型胶原表达。 Objective:To investigate the effect of macrophage-derived TGF-βin proliferation,and reveal the role of macrophage in the process of myelofibrosis.Methods:Mouse bone marrow-derived macrophages(BMMs)and fibroblasts were isolated and cultured;macrophages were activated by LPS and conditioned medium was collected;expression of TGF-βand collagenⅠwere analyzed by ELISA,effects of TGF-βand BMMs coditioned medium on cell proliferation were detected by MTT assay.Results:LPS induced the activation expression of TGF-βin BMMs;TGF-βand macrophages conditioned medium promoted the proliferation and collagenⅠexpression in bone marrow fibroblasts,while anti-TGF-βantibodies treatment inhibited the proliferation and collagenⅠexpression.Conclusion:Macrophage-derived TGF-βcould promote the proliferation of bone marrow fibroblasts and expression of collagenⅠ.
