摘要 目的:建立超高效液相色谱波长切换法同时测定银花感冒颗粒中绿原酸、连翘苷、连翘酯苷A、甘草酸铵、5-O-甲基维斯阿米醇苷5种成分的含量。方法:采用ACQUITY UPLC BEH C18色谱柱(100 mm×2.1 mm,1.7μm),以乙腈(A)-0.1%甲酸溶液(B)为流动相进行梯度洗脱,波长为327 nm(0~3.0 min)、330 nm(3.0~7.0 min)、254 nm(7.0~8.5 min)、277 nm(8.5~10.0 min)、237 nm(10.0~13.0 min),流速为0.4 ml·min^-1,柱温为25℃。结果:绿原酸、连翘苷、连翘酯苷A、甘草酸铵、5-O-甲基维斯阿米醇苷分别在0.009~0.452,0.005~0.234,0.005~0.228,0.013~0.645,0.005~0.276μg(r≥0.9994)范围内线性关系良好,平均加样回收率为99.9%~102.4%,RSD≤1.5(n=6)。结论:建立的UPLC方法操作简单,重复性好,可用于银花感冒颗粒的质量控制。 Objective:To develop a UPLC method for the simultaneous determination of 5 components in Yinhuang Ganmao granules,including 4'-O-beta-glucopyranosyl-5-O-methylvisamminol,chlorogenic acid,forsythoside A,phillyrin and monoammonium glycyrrhizinate.Methods:An ACQUITY UPLC BEH C18 column(100 mm×2.1 mm,1.7μm)was used.The mobile phase was acetonitrile(A)-0.1%formic acid solution(B)with gradient elution at a flow rate of 0.4 ml·min^-1.The detection wavelengths were 327 nm(0-3.0 min),330 nm(3.0-7.0 min),254 nm(7.0-8.5 min),277 nm(8.5-10.0 min)and 237 nm(10.0-13.0 min).The column temperature was set at 25℃.Results:Chlorogenic acid,phillyrin,forsythiaside A,monoammonium glycyrrhizinate and 4'-O-beta-glucopyranosyl-5-O-methylvisamminol had a good linear relationship in the range of 0.009-0.452,0.005-0.234,0.005-0.228,0.013-0.645,0.005-0.276μg(r≥0.9994),respectively.The average recoveries were all within the range of 99.9%-102.4%(RSD≤1.5,n=6).Conclusion:The determination method is convenient,fast and accurate,which can be used for the quality control of Yinhuag Ganmao granules.
