摘要
目的探讨帕金森病(Parkinson's disease,PD)患者肠道嗜黏蛋白阿克曼菌(Akkermansia muciniphila,Akk)各亚型丰度的变化及其临床价值.方法收集我院40例帕金森病患者(PD组)和40例健康体检者(对照组)的粪便标本和血液标本.采用实时荧光定量PCR(RT-qPCR)方法检测粪便中嗜黏蛋白阿克曼菌各亚型丰度的变化;同步分析血清尿酸(UA)、甘油三酯(TG)、总胆固醇(TC)和空腹血糖(Glu)水平.结果PD组和对照组均表现为AkkⅠ的丰度最高,AkkⅢ的丰度最低.在65岁及以上的PD患者肠道中AkkⅡ的丰度明显降低(P<0.05).与对照组相比,PD组的血清尿酸水平明显降低(P<0.05).在PD组中,患者AkkⅠ的丰度与血清尿酸水平呈正相关(P<0.05);患者病程与AkkⅠ的丰度、血清尿酸水平呈负相关(P<0.05).结论PD组中,患者肠道AkkⅠ的丰度与血清尿酸水平呈正相关.PD患者病程与AkkⅠ的丰度和血清尿酸水平呈负相关.联合患者肠道低AkkⅠ丰度和低血清尿酸水平指标,可作为PD早期诊断的新型生物标志物.
Objective To investigate the relationship between abundance changes of Akkermansia muciniphila(Akk)subtypes and Parkinson′s disease(PD)and its clinical value.Methods Fecal samples and blood samples were collected from 40 Parkinson′s disease patients(PD group)and 40 healthy controls(control group)in our hospital.Real⁃time quantitive polymerase chain reaction(RT⁃qPCR)was used to detect the abundance of Akk subtypes in the stool samples.Meanwhile,serum levels of uric acid(UA),triglycerides(TG),total cholesterol(TC),and fasting blood glucose(Glu)were measured.Results In PD group and control group,the abundance of AkkⅠwas the highest,and the abundance of AkkⅢwas the lowest.The abundance of AkkⅡsignificantly reduced in PD patients aged 65 years older(P<0.05).Compared with control group,the serum uric acid level significantly reduced in PD group(P<0.05).In PD group,the abundance of AkkⅠwas positively correlated with the serum UA level(P<0.05).The course of patients with PD was negatively correlated with AkkⅠabundance and the serum UA level(P<0.05).Conclusion In PD patients,the intestinal abundance of AkkⅠwas positively correlated with the serum UA level.The course of PD patients has a negative correlation with AkkⅠabundance and serum UA level.
