摘要
目的观察三七总皂苷对大鼠血瘀证相关指标的影响及治疗作用。方法应用肾上腺素加冰水建立大鼠急性血瘀模型,随机分成模型组、舒血宁组、三七总皂苷低剂量组、高剂量组,连续注射1周后取材。采用LBY-S5血栓形成仪测血栓长度;用LBY-N6A自清洗旋转式黏度计测血液黏度;用试剂盒(冻干型)方法测定凝血酶时间(TT)。结果与模型组比较,注射三七总皂苷的各组大鼠均能够显著抑制体外血栓形成;抑制红细胞压积、降低血液黏度;降低血小板聚集率,延长凝血时间。结论三七总皂苷对大鼠急性血瘀模型血液流变学、血小板黏附及聚集和抗血栓有明显作用,其作用机制与改善微循环、红细胞的变形能力、血液的高凝状态有关。
Objective To observe the changes and therapeutic effects of Panax notoginseng saponins on the related indexes of blood stasis syndrome in rats.Methods Acute blood stasis model of rats was established by using adrenaline plus ice water.They were randomly divided into model group,Shuxuening group,low dose group of Panax notoginseng saponin 10.0 mg·kg^-1,high dose group 20.0 mg·kg-1,and were taken after continuous injection for 1 week.The thrombus length was measured using a LBY-S5 thrombosis instrument;the blood viscosity was measured using a LBY-N6A self-cleaning rotary viscometer;the thrombin time(TT)was determined using a kit(freeze-dried)method.Results Compared with the model group,rats in the group injected with Panax notoginseng saponins significantly inhibited thrombosis in vitro;inhibited hematocrit,decreased blood viscosity,decreased platelet aggregation rate,and prolonged clotting time.Conclusion Panax notoginseng saponins have significant effects on hemorheology,platelet adhesion and aggregation and antithrombotic effects in acute blood stasis model in rats.The mechanism of action is related to the improvement of microcirculation,erythrocyte deformability and hypercoagulability of blood.
