摘要
目的:探讨为膝骨性关节炎大鼠使用壮骨活血汤进行治疗对其血清TNF-α及IL-6水平的影响。方法:将40只无特定病原体(SPF级)的健康雄性SD大鼠随机分为空白组、模型组、低剂量组、高剂量组及对照组。对空白组大鼠不做任何处理。将模型组大鼠、低剂量组大鼠、高剂量组大鼠及对照组大鼠建立成为膝骨性关节炎大鼠模型。为低剂量组大鼠使用低剂量的壮骨活血汤进行治疗。为高剂量组大鼠使用高剂量的壮骨活血汤进行治疗。为对照组大鼠使用盐酸氨基葡萄糖胶囊进行治疗。为模型组大鼠和空白组大鼠均每天喂食适量的生理盐水。然后观察各组大鼠的临床表现、膝关节的疼痛阈值、血清肿瘤坏死因子-α(TNF-α)的水平、血清白细胞介素-6(IL-6)的水平、膝关节软骨组织的形态及膝关节的Makin’s评分。结果:治疗后,空白组大鼠、模型组大鼠、低剂量组大鼠、高剂量组大鼠及对照组大鼠的临床表现及膝关节软骨组织的形态均有所不同。治疗后,低剂量组大鼠、高剂量组大鼠及对照组大鼠膝关节的疼痛阈值均高于模型组大鼠,其血清TNF-α的水平、血清IL-6的水平及膝关节的Makin’s评分均低于模型组大鼠,P<0.05。治疗后,高剂量组大鼠膝关节的疼痛阈值高于对照组大鼠及低剂量组大鼠,其血清IL-6的水平及膝关节的Makin’s评分均低于低剂量组大鼠,其血清TNF-α的水平低于对照组大鼠及低剂量组大鼠,P<0.05。治疗后,低剂量组大鼠血清TNF-α的水平、血清IL-6的水平及膝关节的Makin’s评分均高于对照组大鼠,P<0.05。治疗后,高剂量组大鼠与对照组大鼠血清IL-6的水平及膝关节的Makin’s评分相比,P>0.05。结论:为膝骨性关节炎大鼠使用壮骨活血汤进行治疗可改善其临床症状及膝关节软骨组织的形态,提高其膝关节的疼痛阈值,降低其血清TNF-α及IL-6的水平,减少其膝关节组织学的改变。与使用低剂量壮骨活血汤相比,为膝骨性关节炎大鼠使用高剂量壮骨活血汤进行治疗可获得更好的效果。
Objective:To explore the therapeutic effect and partial mechanism of Zhuanggu Huoxue Decoction on knee osteoarthritis animal model.Methods:A rat model of knee osteoarthritis was established by intra-articular injection of 2%papain.The effects of Zhuanggu Huoxue Decoction(ZHXD)on knee arthritis in rats were observed with the general condition,pain threshold,joint pathological section and Makin´s score of bone and joint as indicators.The contents of TNF-alpha and IL-6 in serum of rats were detected by ELISA,and the mechanism of Zhuanggu Huoxue Decoction in treating knee osteoarthritis was preliminarily discussed.Results:Compared with the model group,each group of Zhuanggu Huoxue Decoction could increase the pain threshold,improve the general condition and decrease the Makin´s score,and significantly reduce the contents of TNF-alpha and IL-6 in the serum of rats.Conclusion:Zhuanggu Huoxue Decoction has therapeutic effect on knee osteoarthritis rats,and its mechanism may be related to its inhibition of TNF-alpha and IL-6 secretion.
