一种高寒草地土壤微生物生物量磷的测定方法 被引量:2

摘要 磷是微生物生长的必须营养元素,也是微生物细胞成分的组成元素。土壤微生物生物量磷作为土壤微生物生物量中的一部分,其对环境的变化非常敏感。因此准确测定土壤中微生物生物量磷的含量对检测土壤环境及其土壤养分具有重要意义。本文采用硫酸过硫酸钾消化,高温高压湿热处理对高寒草地土壤微生物生物量磷含量测定方法进行了研究,结果显示:25mL的土壤碳酸氢钠提取液,依次经1mL33%硫酸和4mL5%过硫酸钾消化后,110℃~115℃处理25min或120℃处理10min测得的微生物磷与文献报道的土壤微生物磷含量范围吻合,该法可为高寒草地土壤微生物生物量磷的进一步研究提供一种简单、快捷的的测定方法,减少实验过程中磷的损失。 Phosphorus is not only a necessary nutrient element for microbial growth,but also a constituent element of microbial cell components.Soil microbial biomass phosphorus,as a part of soil microbial biomass,is very sensitive to environmental changes.Therefore,the accurate determination of microbial biomass phosphorus content in soil is of great significance for the detection of soil environment and soil nutrients.In this paper,the determination method of soil microbial biomass phosphorus content in alpine grassland was studied by sulfuric acid-potassium persulfate digestion,and high temperature and high pressure,humid-heat treatment.The results showed that the soil sodium bicarbonate extract of 25ml was digested by 1ml,33%sulfuric acid and 4ml,5%potassium persulfate,and treatment at 110℃~115℃for 25min or 120℃for 10min,the microbial phosphorus measured by this method was consistent with the range of soil microbial P content reported in the literature.This method can provide a simple and rapid method for the further study of soil microbial biomass phosphorus in alpine grassl,and and reduce the loss of phosphorus in the process of experiment.
