摘要
目的探讨细胞因子白细胞介素17E(IL-17E)及其受体白细胞介素17RB(IL-17RB)、细胞因子白细胞介素17F(IL-17F)及其受体白介素17RC(IL-17RC)在正常前列腺组织(normal prostate,NP)、良性前列腺增生组织(benign prostatic hyperplasia,BPH)和前列腺癌(prostate cancer,Pca)组织中的表达差异.方法利用免疫组织化学染色法检测了6例NP、39例BPH和20例Pca标本中IL-17E、IL-17RB、IL-17F和IL-17RC阳性表达率,并分析配体与相应受体阳性表达率的相关性,配体阳性表达率与前列腺癌恶性程度的关系.结果在前列腺组织中,IL-17E、IL-17RB、IL-17F和IL-17RC主要表达于腺上皮细胞以及单个核细胞,少部分表达于血管内皮细胞及部分癌细胞.与BPH和NP比较,在Pca组织中,IL-17E表达明显降低,差异具有统计学意义(P<0.05),且随着前列腺癌Gleason分级增加,IL-17E的表达降低;与NP比较,IL-17RB在Pca中表达增加,差异具有统计学意义(P<0.01);与BPH和NP比较,IL-17F在Pca组织中表达明显增加,差异具有统计学意义(P<0.05),随着前列腺癌Gleason分级的增加,IL-17F表达增强;与BPH和NP比较,IL-17RC在Pca中表达也增加,差异具有统计学意义(P<0.05,P<0.01);在Pca组织中,IL-17F与IL-17RC表达呈正相关性(r=0.633,P=0.021).结论 IL-17F可能具有促进前列腺癌发生发展的效应,而且主要是通过IL-17F-IL-17RC信号通路的激活实现的.
Objective To explore expression difference of cytokine interleukin-17E(IL-17E)and its receptor interleukin-17RB(IL-17RB),interleukin-17F(IL-17F)and its receptor interleukin-17RC(IL-17RC)in normal prostate(NP),benign prostatic hyperplasia(BPH)and prostate cancer(Pca).Method Immunohistochemistry staining assay was applied to detect IL-17E,IL-17RB,IL-17F and IL-17RC expression in 6 NP specimens,39 BPH specimens and 20 Pca specimens.Spss10 software was applied to analyze the relationship between relative ligands with their respective receptors and ligands with malignancy of cancer.Results IL-17E,IL-17RB,IL-17F and IL-17RC mainly expressed glandular epithelium and mononuclear cells,and small part in vascular endothelial cells as well as partial cancerous cells.Compared with BPH and NP,IL-17E expression obviously decreased in Pca,the difference was statistically significant(P<0.05).With the increase of Gleason grade,IL-17E expression reduced obviously.Compared with BPH and NP,IL-17RB had obviously increased in Pca,the difference was statistically significant(P<0.01).Compared with BPH and NP,IL-17F elevated obviously in Pca,the difference was statistically significant(P<0.05).With the increase of Gleason grade,IL-17F expression increased more evident.Compared with BPH and NP,IL-17RC showed more significant in Pca,the difference was statistically significant(P<0.01).Positive correlation between IL-17F and IL-17RC expression in Pca tissues was found(r=0.633,P=0.021).Conclusion The expression of IL-17F increased significantly in Pca and its expression increased obviously with the increase of the malignancy of Pca.IL-17F has protumor effect in occurrence and development with the IL-17F-IL-17RC signal activation.
