摘要
目的研究蓟黄素对心力衰竭(HF)大鼠心肌细胞自噬水平的影响及作用机制。方法将造模成功HF大鼠随机分为5组:模型组、对照组和低、中、高3个剂量组,每组15只。术后第5周开始给药,对照组灌胃给予10 mg·kg^-1普萘洛尔;3个剂量实验组灌胃给予蓟黄素溶液25,50,100 mg·kg^-1(药品均加0.9%NaCl配制成1 mg·mL^-1溶液),1次/天,连续给药4周。用高分辨率小动物超声系统检测左室射血分数(LVEF),用蛋白质印迹法检测各组微管相关蛋白1轻链3(LC3)表达情况。结果模型组、对照组和低、高2个剂量实验组的LVEF分别为(39.42±9.12)%,(62.43±7.23)%,(43.51±10.42)%和(64.31±8.42)%,对照组和高剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。模型组、对照组和低、高2个剂量实验组的双层膜自噬体数量(23.62±3.52),(8.16±1.62),(18.47±3.65)和(7.64±1.52)个;这4组的LC3水平分别为(87.69±10.54)%,(41.62±7.44)%,(67.52±6.88)%和(42.51±5.62)%;这4组的LC3Ⅱ/LC3Ⅰ比值分别为2.33±0.61,2.02±0.58,1.76±0.52和1.52±0.43。上述指标:对照组和低、高2个剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论蓟黄素能显著降低HF大鼠的过度自噬水平,提高心功能。
Objective To investigate the effect and its mechanism of cirsimaritin on autophagy level in rats with heart failure.Methods After the establishment of heart failure model,rats were divided into five groups:model group,control group,experimental-L group,experimental-M group and experimental-H group,each with 15 rats.Rats in control group was given 10 mg·kg^-1 propranolol;rats in experimental-L,-M,-H groups were given 25,50,100 mg·kg^-1 cirsimaritin.At the 5th week after operation,left ventricular ejection fraction(LVEF)were detected by high resolution ultrasound system for small animals;the levels of the relative proteins of light chain 3(LC3)were detected by Western blot.Results LVEF in model group,control group,experimental-L group and experimental-H group were(39.42±9.12)%,(62.43±7.23)%,(43.51±10.42)%and(64.31±8.42)%.Comparison between control group,experimental-H group and model group,the differences were significant(all P<0.05).The number of double membrane autophagy in the four groups were 23.62±3.52,8.16±1.62,18.47±3.65 and 7.64±1.52;LC3 levels in the four groups were(87.69±10.54)%,(41.62±7.44)%,(67.52±6.88)%and(42.51±5.62)%;ratio of LC3II/LC3I in the four groups were 2.33±0.61,2.02±0.58,1.76±0.52 and 1.52±0.43.Comparison between control group,experimental-L group,experimental-H group and model group,the differences of the factors were significant(all P<0.05).Conclusion Cirsimaritin can significantly reduce the level of excessive autophagy and ameliorate the cardiac function of rats with heart failure.
