长链非编码RNA MALAT1对鼻咽癌转移和上皮间质转化的影响 被引量:1

Effects of long non-coding RNA MALAT1 on metastasis and epithelial mesenchymal transformation of nasopharyngeal carcinoma

摘要 目的探讨长链非编码RNA MALAT1(Lnc RNA MALAT1)通过蛋白酪氨酸激酶2/信号转导子与激活子3(JAK2/STAT3)信号通路调控鼻咽癌上皮间质转化(EMT)的机制。方法将鼻咽癌细胞SUNE-1分为3组:空白组、第一转染组和第二转染组。第一转染组和第二转染组分别转染NC片段和siMALAT1转染干扰序列,空白组细胞不做任何处理。Transwell实验检测细胞侵袭能力,蛋白质印迹法检测E-钙黏蛋白、波形纤维蛋白、JAK2和STAT3蛋白的相对表达水平。结果空白组、第一转染组和第二转染组的细胞侵袭数分别为(441.63±20.65),(438.25±19.03)和(197.75±8.31)个;这3组的E-钙黏蛋白相对表达量分别为0.99±0.08,0.94±0.08和1.68±0.04;这3组的波形纤维蛋白相对表达量分别为1.00±0.06,0.97±0.07和0.47±0.02;这3组的JAK2蛋白相对表达量分别为0.97±0.09,0.94±0.07和0.29±0.04;这3组的STAT3蛋白相对表达量分别为0.95±0.05,0.97±0.08和0.43±0.03。第二转染组与空白组和第一转染组比较,细胞侵袭数、波形纤维蛋白、JAK2和STAT3相对表达量均明显下降,E-钙黏蛋白相对表达量明显升高,差异均有统计学意义(均P<0.05)。结论 Lnc RNA MALAT1在鼻咽癌组织及鼻咽癌细胞中高表达,高表达Lnc RNA MALAT1可能通过强化JAK2/STAT3信号通路促进EMT过程,从而影响鼻咽癌的发展。 Objective To investigate the mechanism of long non-coding RNA MALAT1(Lnc RNA MALAT1)regulating the epithelial mesenchymal transition(EMT)of nasopharyngeal carcinoma through protein tyrosine kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3)signal pathway.Methods The nasopharyngeal carcinoma SUNE-1 cells were divided into three groups:Blank group,and transfection-1,transfection-2 groups.SUNE-1 cells in blank group were not transfected;SUNE-1 cells in transfection-1 group and transfection-2 group were transfected control negative and siMALAT1,respectively.Transwell assay was used to detect cell invasion ability(OD value).Western blot was used to detect the expressions of E-cadherin,Vimentin,JAK2 and STAT3.Results The invasion number of cells in blank group,transfection-1,and transfection-2 groups were(441.63±20.65),(438.25±19.03)and(197.75±8.31),respectively;the relative expression levels of E-cadherin were 0.99±0.08,0.94±0.08 and 1.68±0.04,respectively;the relative expression levels of Vimentin were 1.00±0.06,0.97±0.07 and 0.47±0.02,respectively;the relative expressions of JAK2 protein were 0.97±0.09,0.94±0.07 and 0.29±0.04,respectively;the relative expressions of STAT3 protein were 0.95±0.05,0.97±0.08 and 0.43±0.03,respectively.Compared with blank group and transfection-1 group,the relative expressions of Vimentin,JAK2 and STAT3 in transfection-2 group were decreased significantly,while the relative expressions of E-cadherin were increased significantly(all P<0.05).Conclusion Lnc RNA MALAT1 is highly expressed in nasopharyngeal carcinoma tissues and nasopharyngeal carcinoma cells.The highly expressed Lnc RNA MALAT1 may promote the EMT process by strengthening the JAK2/STAT3 signaling pathway,thus affecting the development of nasopharyngeal carcinoma.
