龟鹿二仙胶对去势大鼠骨骼肌Akt蛋白表达的影响

Influence of Guilu ErXian Glue on the Expression of Akt Protein in Sketel Muscle of Ovariectomy Rats

目的:探讨龟鹿二仙胶对去势大鼠骨骼肌Akt蛋白表达的影响。方法:40只6月龄SD大鼠随机分为假手术、模型组、龟鹿二仙胶组、雌激素组各10只,假手术组、模型组灌胃生理盐水,龟鹿二仙胶组给予龟鹿二仙胶颗粒剂灌胃,雌激素组给予戊酸雌二醇。治疗12周后活体骨密度(BMD)检测,同时检测骨骼肌Akt蛋白、mRNA表达。结果:模型组BMD明显低于假手术组(P<0.05),龟鹿二仙胶组和雌激素组高于模型组(P<0.05);模型组蛋白Akt、p-Akt表达量明显低于假手术组(P<0.05);而龟鹿二仙胶组和雌激素组高于模型组(P<0.05);模型组Akt1 mRNA表达量明显低于假手术组(P<0.05),而龟鹿二仙胶组和雌激素组明显高于模型组(P<0.05)。结论:龟鹿二仙胶对去势大鼠有明显的治疗作用,能改善BMD,提高骨骼肌Akt蛋白和基因的表达量。

Objective:To investigate the role of Guilu ErXian Glue on the Expression of Akt protein in skeletal muscle of ovariectomy rats.Methods:6-month-old rats were randomly divided into Sham,model,GuiLu ErXian,Estrogen groups,10 rats each group.The Sham and model group were given normal saline,the GuiLu ErXian group and Estrogen Treatment group were given responding drugs.After 12 weeks treatment,we measured the bone mineral density(BMD)of the rats and the detection of Akt protein and mRNA expression in skeletal muscle.Results:The BMD of model group rats were significantly lower than Sham group(P<0.05);GuiLu ErXian group and Estrogen group are higher than model group(P<0.05).The expression of protein Akt and p-Akt in model group rats is significantly lower in Sham group(P<0.05);GuiLu ErXian group and Estrogen group are higher than model group(P<0.05).The expression of protein Akt1 mRNA in model group rats is significantly lower than in Sham group(P<0.05);GuiLu ErXian Glue group and Estrogen group are higher than model group(P<0.05).Conclusion:GuiLu ErXian Glue has an obvious treatment on ovariectomy rats,which can improve the BMD of rats and increase the expression of Akt protein and mRNA in skeletal muscle.
