摘要
目的观察不完全典型的鼠疫耶尔森菌菌落形态,为鼠疫常规监测或疫情判定提供基础资料,避免漏检。方法 2017年冬季到次年春季,在河北省康保县鼠疫疫源地发生了动物间鼠疫流行,共检出鼠疫耶尔森菌6株,采用经典的"4步检验"法对6株鼠疫耶尔森菌进行试验,观察其在亚硫酸钠琼脂培养基上的生长情况和菌落形态特征。结果 2017-2018年河北省康保县鼠疫疫源地新分离的6株菌在亚硫酸钠琼脂培养基上28℃培养24 h,发育良好,光学显微镜下观察菌落中央为黄褐色粗糙颗粒,均未看到鼠疫菌的典型菌落形态,即周围有一圈宽窄不等,呈锯齿状、薄而透明的花边。结论这种不完全典型的鼠疫耶尔森菌菌落形态在常规监测或疫情判定过程中应引起高度重视,避免漏检,为鼠疫监测和防治提供基础资料。
Objective To observe the colony morphology of Yersinia pestis which was not completely typical,providing basic information for routine monitoring of plague or disease determination to avoid missed detection.Methods From the winter of 2017 to the spring of 2018,the plague epidemic among animals occurred in the plague foci of Kangbao County,Hebei Province.Six strains of Y.pestis were isolated.The six strains of Y.pestis were tested by the classical“four-step test”method.The growth and colony morphology of Y.pestis on sodium sulfite agar medium were observed.Results The newly isolated 6 strains of bacteria from plague foci of Kangbo County,Hebei Province in 2017 and 2018 were cultured at 28℃for 24 h on sodium sulfite AGAR medium,showing good development.Under the light microscope,the center of the colony was observed as yellow-brown coarse particles.No typical colony morphology of Y.pestis was observed.The shape of the colony with a jagged,thin and transparent lace around the circle was not seen.Conclusion This kind of incompletely typical colony morphology of Y.pestis should be paid great attention on routine surveillance or epidemic situation determination,so as to avoid missed detection and provide basic information for the monitoring and control of plague.
