维替泊芬对异丙肾上腺素诱导小鼠心肌纤维化的影响 被引量:1

Effects of verteporfin on cardiac function in mice with isoproterenol-induced myocardial fibrosis

摘要 目的探讨维替泊芬(VP)对异丙肾上腺素(ISO)诱导的小鼠心肌纤维化是否有抑制作用及对心功能的影响。方法 6周龄C57BL/6小鼠(购自中山大学实验动物中心)随机分为4组。对照组予以尾静脉注射1%二甲基亚砜(DMSO);ISO组予以皮下注射ISO (每天20 mg/kg);VP组予以尾静脉注射VP(每天2 mg/kg);ISO+VP组分别予以皮下注射ISO(每天20 mg/kg)和尾静脉注射VP(每天2 mg/kg)。连续给药7 d后行超声心动图检查评估心脏形态及功能。取材后行心脏质量、心脏体重比测定,天狼猩红染色检测心肌纤维化变化情况。酶联免疫吸附测定(ELISA)血浆氨基末端脑钠素原(NT-pro BNP)浓度。组间比较采用单因素方差分析。结果成功建立了ISO诱导小鼠心脏纤维化模型。ISO组小鼠的左心室收缩末内径[(2.20±0.63) mm比(1.44±0.45) mm,F=9.730,P<0.05]及容积[(18.33±12.17) μl比(6.32±5.04) μl,F=8.890,P<0.05]较ISO+VP组增大,差异有统计学意义,左心射血分数(EF)较ISO+VP组降低[(56.65±11.41)%比(82.35±10.30)%,F=15.670,P<0.05],差异有统计学意义;ISO组小鼠的心脏质量[(0.11±0.01) mg比(0.10±0.01) mg,F=13.560,P<0.05]及心脏体重指数[(0.57±0.03)%比(0.51±0.02)%,F=23.950,P<0.05]均大于ISO+ VP组,差异有统计学意义;ISO组小鼠血浆NT-pro BNP浓度较VP组升高[(0.57±0.16) μg/L比(0.23±0.07) μg/L,F=9.340,P<0.05],差异有统计学意义;天狼猩红染色结果显示ISO组小鼠左室心肌纤维化较ISO+VP组严重[(17.48±5.85)%比(9.27±2.62)%,F=30.190,P<0.05],差异有统计学意义。结论 VP可抑制ISO诱导的心脏纤维化并改善左心收缩功能。 Objective To investigate the inhibitory effect of Verteporfin(VP)on myocardial fibrosis induced by isoproterenol(ISO)in mice and its effect on cardiac function.Methods The 6-week-old C57BL/6 mice supplied by the Laboratory Animal Center of Sun Yat-sen University were randomly divided into 4 groups.In the control group,1%dimethyl sulfoxide(DMSO)was injected into the tail vein.ISO group received subcutaneous injection of ISO.VP was injected into the tail vein in VP group.The ISO+VP group received subcutaneous injection of ISO and caudal vein injection of VP.Echocardiography was performed 7 days after continuous administration to assess cardiac morphology and function.After sampling,heart mass,heart weight ratio and Sirius red were measured.Plasma NT-pro BNP concentration was determined by enzyme linked immunosorbent assay(ELISA).One-way anova was used for inter-group comparisons.Results A model of ISO-induced cardiac fibrosis in mice was established.Compared with ISO+VP group,the final diameter[(2.20±0.63)mm vs.(1.44±0.45)mm,F=9.730,P<0.05]and volume of left ventricular contraction[(18.33±12.17)μl vs.(6.32±5.04)μl,F=8.890,P<0.05]in ISO group were both increased,and the left ventricular ejection fraction(EF)was decreased[(56.65±11.41)%vs.(82.35±10.30)%,F=15.670,P<0.05].The heart mass[(0.11±0.01)mg vs.(0.10±0.01)mg,F=13.560,P<0.05]and body mass index[(0.57±0.03)%vs.(0.51±0.02)%,F=23.950,P<0.05]of mice in ISO group were significantly increased as compared with those in VP+ISO group.Plasma nt-pro BNP concentration in mice in ISO group was higher than that in VP group[(0.57±0.16)μg/L vs.(0.23±0.07)μg/L,F=9.340,P<0.05].The left ventricular fibrosis in ISO group was more severe than that in ISO+VP group[(17.48±5.85)%vs.(9.27±2.62)%,F=30.190,P<0.05].Conclusion VP inhibits ISO-induced cardiac fibrosis and improves left ventricular systolic function.
