摘要
随着生活水平的提高,2型糖尿病、非酒精性脂肪肝病等已成为全球主要的公共卫生健康问题。肝脏的胰岛素信号在调控肝脏的糖原合成、糖异生、糖酵解、脂质的合成和分解等多种代谢过程中起到关键作用,而肝脏胰岛素抵抗将导致明显的糖脂代谢紊乱,是2型糖尿病及非酒精性脂肪肝病等代谢相关疾病发生发展极其重要的因素。然而,目前肝脏胰岛素抵抗的发生机制尚未完全明了,因此,深入研究肝脏胰岛素抵抗的相关机制对相关疾病的防治有着重大意义。本文旨在阐明肝脏胰岛素抵抗相关机制研究进展。
With the lifestyle transitions,metabolic diseases such as type 2 diabetic mellitus(T2DM)and non-alcoholic fatty liver disease(NAFLD)have become the major global public health problems.Insulin signaling pathway in the liver plays a substantial role in regulating various metabolic processes including hepatic glycogen synthesis,gluconeogenesis,glycolysis,lipogenesis and lipolysis.Hepatic insulin resistance disturbs the homeostasis of glycolipid metabolism,playing an essential role in the development of T2DM,NAFLD and other metabolic diseases.However,the pathogenesis of hepatic insulin resistance has not yet been fully understood.Thus,the study on the mechanism of hepatic insulin resistance is of great significance for the prevention and treatment of metabolic disorders.This review aims to elucidate the potential mechanisms of hepatic insulin resistance.
