摘要
目的:探讨miR-1233和HoxB3在复发性流产(RM)患者及人工流产患者绒毛组织中表达水平的差异以及其对血浆血管内皮生长因子(VEGF)的影响。方法:选择2017年3月—2019年3月在妇产科就诊的RM患者为观察组,共90例,分为观察1组(自然流产2次)、观察2组(自然流产3次)和观察3组(自然流产≥4次),每组30例。选择同期意外妊娠行人工流产的30例患者为对照组(无自然流产史及其他不良妊娠史且非人工受孕等)。用逆转录-聚合酶链反应(RT-PCR)检测两组患者绒毛组织中miR-1233,HoxB3的mRNA表达水平。用酶联免疫法(ELISA)分析各组血清中VEGF的差异。结果:与对照组比较,观察1,2,3组绒毛中miR-1233的表达明显增高,HoxB3和VEGF的表达明显降低,差异有统计学意义(P<0.05)。在观察1,2,3组间比较,可以看出随着流产次数的增加,绒毛中miR-1233的表达逐渐增高,HoxB3和VEGF的表达越来越低,差异有统计学意义(P<0.05)。结论:推测在复发性流产绒毛中高表达的miR-1233,一方面可加速滋养细胞的凋亡,另一方面通过抑制HoxB3的表达,影响VEGF的合成分泌,从而减慢绒毛组织细胞的增殖分化,加快胚胎组织凋亡,最终导致妊娠失败。
Objective:To investigate the expression of miR-1233 and HoxB3 in the villi of patients with recurrent abortion(RM)and induced abortion,and the effect of miR-1233 and HoxB3 on vascular endothelial growth factor(VEGF).Methods:From March 2017 to March 2019,30 RM patients in our hospital were selected as the experimental group,which were divided into experimental group 1(2 spontaneous abortions),experimental group 2(3 spontaneous abortions),experimental group 3(30 spontaneous abortions≥4).Thirty cases of induced abortion in the same period were randomly selected as the control group.The mRNA expression levels of miR-1233 and HoxB3 in the villi of the two groups were detected by RT-PCR,and the differences of VEGF in serum were analyzed by enzyme linked immunosorbent assay(ELISA).Results:Compared with the control group,the expression of miR-1233 in the villi of exper-imental group 1,2 and 3 was significantly higher,and the expression of HoxB3 and VEGF was significantly lower(P<0.05).The expression of miR-1233 and HoxB3 and VEGF in the villi increased with the increase of abortion frequency,and the difference was statistically significant(P<0.05).Conclusion:We speculate that the high expression of miR-1233 in the villi of recurrent abortion can accelerate the apoptosis of trophoblasts on the one hand,and on the other hand,it can affect the synthesis and secretion of VEGF by inhibiting the expression of HoxB3,thus slowing down the proliferation and differentiation of villi cells,showing the accelerated apoptosis of fetal tissues,leading to pregnancy failure.
