单宁对玉木耳菌丝生长发育的影响 被引量:1

Effect of Tannin Concentration on Mycelial Growth and Development of Auricularia cornea

摘要 为探究利用单宁含量较高的针叶林木屑栽培玉木耳(Auricularia cornea)的可能性,通过设置不同单宁浓度固体及液体培养基培养玉木耳菌丝,测定了菌丝的长势、生长速度、菌丝体的生物量、锁状联合比率以及菌丝显微形态特征,研究了不同浓度的单宁对玉木耳菌丝生长发育的影响。结果表明:在液体摇瓶实验中随着单宁浓度的升高,菌丝体生物量呈显著负相关关系(P<0.05),单宁质量浓度为0.05 g/L时,菌丝体干质量与对照(0 g/L)相比减少了32.6%,当单宁质量浓度为0.25~0.30 g/L时,分别减少71.8%和77.7%。在平板实验中单宁质量浓度在0.05~0.10 g/L时,菌丝生长速度、锁状联合比率和菌丝体分支率显著增加(P<0.05),当单宁质量浓度达到0.10 g/L时,相较于对照(0 g/L)分别提高了15.1%、13.2%和13.9%。随着单宁浓度的升高,菌丝生长速度、锁状联合比率和菌丝体分支率均呈现不同程度下降。实验表明低浓度单宁促进菌丝生长,为利用针叶林木屑栽培玉木耳提供了理论基础。 With different concentrations of tannin on the plate and liquid shake flask of Auricularia cornea,the mycelial growth rate,mycelial dry-weight,clamp-connection hyphae ratio,and the mycelial branching frequency were determined.In the experiment,the dry weight of mycelium decreased with the increased tannin concentration.When tannin concentration was 0.05 g/L,the dry weight of mycelium decreased by 32.6%in comparison with control(0 g/L).When the tannin concentration was increased from 0.25 to 0.30 g/L,the dry weight of mycelium significantly decreased from 71.8%to 77.7%,respectively,in comparison with control(0 g/L).In the PDA medium experiment,when the concentration of tannin is between 0.05 g/L and 0.10 g/L,the mycelial growth rate,clamp-connection hyphae ratio,and mycelial branching frequency were increased compared with the control group(0 g/L).When the tannin concentration reaches 0.10 g/L,the ratios were increased by 15.1%,13.2%,and 13.9%,respectively,compared with the control group.With the increase of tannin concentration,the mycelial growth rate,clamp-connection hyphae ratio,and mycelial branching frequency were decreased to different extent.In this study,we clarified the effects of tannin on sawdust on mycelia of the auricularia corneas,and its mechanism of action.The possibility of utilizing coniferous sawdust to cultivate auricularia corneas is also explored,and there is potential for coniferous sawdust used in other edible fungi.
