摘要
目的探究长链非编码RNA LINP1在肝癌组织中的表达及其对肝癌细胞增殖及侵袭转移能力的影响。方法使用qRT-PCR方法检测60例肝癌组织以及癌旁组织、肝癌细胞株及正常肝细胞中LINP1的表达情况。si-LINP1转染肝癌细胞敲低LINP1表达;CCK-8实验检测si-LINP1对肝癌细胞增殖影响;Western blot检测Wnt/β-catenin信号通路蛋白β-catenin和cyclinD1表达。结果LINP1在肝癌组织中的表达水平高于癌旁组织,肝癌细胞高于正常肝细胞系Lo2(P<0.05);敲低LINP1抑制肝癌细胞增殖及侵袭转移,抑制Wnt/β-catenin信号通路蛋白β-catenin和cyclinD1表达(P<0.05)。结论敲低LINP1抑制肝癌细胞的增殖、侵袭和转移与抑制Wnt/β-catenin信号通路相关。
Objective To investigate the expression of long non-coding RNA LINP1 in hepatocellular carcinoma(HCC)and its effect on proliferation,invasion and metastasis of liver cancer cell lines.Methods qRT-PCR assay was used todetect LINP1 mRNA expression in HCC tissues and adjacent tissues of 60 patients,and in the HCC cells lines and normal liver cells.LINP1 expression was knocked down by si-LINP1 transfection.CCk-8 assay was used to detect the effect of si-LINP1 on the proliferation.Western blotting was used to detect the expression of the Wnt/β-catenin signaling pathway proteinβ-catenin and cyclinD1.Results The expression of LINP1 in liver cancer tissues was higher than that in adjacent tissues(P<0.05).LINP1 expression in HCC cell lines was higher than that in normal liver cell line Lo2(P<0.05).LINP1 knockdown inhibited the proliferation,invasion and metastasis(P<0.05).LINP1 knockdown inhibited the expression of Wnt/β-catenin signaling pathway proteinβ-catenin and cyclinD1(P<0.05).Conclusion The expression of LINP1 in liver cancer is elevated,and LINP1 can mediate the Wnt/β-catenin signaling pathway to inhibit the proliferation,invasion and metastasis in HCC.
