miR-21在白藜芦醇诱导A549肺癌细胞株凋亡中的作用

The role of miR-21 in Resveratrol-induced apoptosis of A549 lung cancer cell line

摘要 目的探讨白藜芦醇对A549肺癌细胞株凋亡的影响,并揭示miR-21参与其中的机制。方法以高糖DMEM培养基培养A549肺癌细胞株,给予100μmol/L的白藜芦醇,过表达(或不过表达)miR-21培养3 d。Real-time PCR检测miR-21表达;MTT法检测细胞活力;Hoechst染色观察细胞凋亡;Western blot检测细胞凋亡因子4(programmed cell death 4,PDCD4)的表达。结果白藜芦醇能抑制A549肺癌细胞株miR-21的合成,上调PDCD4的表达,抑制细胞生长,降低细胞活力,诱导A549细胞凋亡(P<0.05)。过表达miR-21能显著抑制白藜芦醇的抗肿瘤作用,PDCD4表达变化不明显,细胞生长状态、活力以及凋亡等均无明显变化(P>0.05)。结论白藜芦醇能通过抑制miR-21合成,降低PDCD4表达,诱导A549肺癌细胞株凋亡。 Objective To explore the effect of Resveratrol on the apoptosis of A549 lung cancer cell line,and to investigate the potential underlying role of microRNA-21.Methods A549 lung cancer cells were cultured in high glucose DMEM medium.Resveratrol(with or without over-expressed microRNA-21)was given at 100μmol/L for 3 days.Real-time PCR was used to detect the expression of microRNA-21,MTT was used to detect cell viability,Hoechst staining was used to observe cell apoptosis;Western blot was used to detect the expression of programmed cell death 4(PDCD4).Results Resveratrol inhibited the synthesis of microRNA-21,together with up-regulation of PDCD4 expression level,inhibition of cell growth,reduction of cell viability and apoptosis of A549 lung cancer cells(P<0.05).The overexpression of microRNA-21 significantly inhibited the anti-tumor effect of Resveratrol,but the expression of PDCD4 had no significant change,and the growth status,viability and apoptosis of cells showd no significant alteration(P>0.05).Conclusion Resveratrol induces apoptosis of A549 lung cancer cell line via inhibiting the synthesis of microRNA-21 and decreasing the expression of PDCD4.
