不同来源鸡血藤化学特征与抗肿瘤作用差异研究 被引量:6

The Difference of Chemical Characteristics and Anti-tumor Effect of Different Sources of Ji Xueteng

摘要 目的:评价不同来源鸡血藤生物活性、化学特征差异性,探讨正品鸡血藤优势,为鸡血藤的质量评价体系完善提供实验基础。方法:利用UPLC建立不同来源鸡血藤乙酸乙酯部位指纹图谱;利用香草醛-盐酸法、分光光度法及MTS法分别测定鸡血藤粗提物原花青素含量、DPPH·自由基清除率及人乳腺癌细胞(MDA-MB-231)IC50。结果:指纹图谱相似度、DPPH·自由基清除率及原花青素含量结果在不同来源鸡血藤之间具有显著差异;UPLC指纹图谱相似度可区分广西鸡血藤、越南鸡血藤、云南鸡血藤与四川鸡血藤,DPPH·清除率及原花青素含量可区分广西鸡血藤、越南鸡血藤、云南鸡血藤与四川鸡血藤;相关性分析结果表明,DPPH·自由基清除率与原花青素含量呈明显正显著相关,自由基清除能力与抑制肿瘤细胞活力呈正显著相关(P<0.05或P<0.01)。结论:正品来源鸡血藤无论是在提取率、原花青素含量还是抑制乳腺肿瘤细胞活力都存在明显优势,原花青素含量可能是区分不同基源鸡血藤抗肿瘤作用的重要指标。 Objective: To evaluate the biological activity and chemical characteristics of different species of Ji Xueteng, and to explore the advantages of Spatholobus suberectus Dunn, and provide experimental basis for the improvement of the quality evaluation system of Spatholobus suberectus Dunn.Methods: The fingerprints of ethyl acetate extracts from different sources were established by UPLC. The vanillin-hydrochloric acid method, spectrophotometry and MTS method were used to determine the proanthocyanidin contents, DPPH·radical clearance rate and human breast cancer cell(MDA-MB-231) IC50 of crude extracts from Ji Xueteng. Results:The Difference results showed that the fingerprint similarity, Clearance ability of DPPH·and Procyanidins content were significantly different among the groups. UPLC fingerprint similarity distinguished among G(Guangxi Ji Xueteng), V(Vietnam Ji Xueteng), Y(Yunnan Ji Xueteng) and S(Sichuan Ji Xueteng). Clearance ability of DPPH·and Procyanidins content distinguished G, V, Y and S. Correlation analysis results showed that Clearance ability of DPPH·was positively correlated with original cyanine contents, Clearance ability of DPPH·and tumor Cell viability inhibition were positively correlated(P <0.01). Conclusion: The authentic source of Spatholobus suberectus Dunn has obvious advantages in the extraction rate, the Procyanidins content and the inhibition of breast tumor cell viability. Procyanidins content might be an important indicator to distinguish anti-tumor activity of different base sources of Ji Xueteng.
