免疫性血小板减少症气不摄血证小鼠模型的动态研究

Dynamic Study of Immune Thrombocytopenia Mice Model with Qibushexue Syndrome

摘要 目的:从证候与疾病两方面观察免疫性血小板减少症(immune thrombocytopenia,ITP)气不摄血证小鼠模型的动态变化特征,初步阐释ITP气不摄血证的形成过程。方法:Balb/c雄性小鼠随机分为正常对照组和模型对照组,模型对照组采用第1 w睡眠剥夺,第2、3 w睡眠剥夺复合腹腔注射抗血小板血清(anti-platelet serum,APS)的方法复制ITP气不摄血证小鼠模型。实验第7 d、10 d、21 d时各取一批小鼠检测体质量、抓力、脉搏幅度等相关证候指标,以及出血、外周血小板(platelet,PLT)计数、脾脏指数、脾脏病理等相关疾病指标。结果:与正常对照组比较,7 d、10 d、21 d时模型对照组小鼠的证候指标(体质量、抓力、脉搏幅度)均显著降低(P<0.01);7 d时模型对照组小鼠无出血情况,脾脏指数无明显变化(P>0.05),10 d、21 d时模型对照组小鼠腹部、尾等不同部位均有不同程度的出血现象,脾脏指数显著增高(P<0.01);7 d、10 d、21 d时模型对照组小鼠PLT计数均显著降低(P<0.01)。结论:睡眠剥夺复合腹腔注射APS法复制的ITP气不摄血证小鼠模型呈现气虚-出血-气虚-气不摄血的动态变化特征。 Objective:To observe the dynamic changes of immune thrombocytopenia(ITP)mouse model with Qibushexue syndrome,and to preliminarily explain the formation process of ITP model with Qibushexue syndrome.Methods:Balb/c mice were randomly divided into the control group and the model group.Mice in the model group were subjected to sleep deprivation in the first week and the sleep deprivation combined intraperitoneal injection of anti-platelet serum(APS)in the second and third weeks,to establish the ITP mice model with Qibushexue syndrome.On the 7 th,10 th and 21 st day of the experiment,a batch of mice were taken to detect related symptom index such as the body mass,the holding power,the pulse amplitude,and disease indicators such as the bleeding grade,PLT count in the peripheral blood,the spleen index,the spleen pathology.Results:Compared with the control group,syndrome indexes such as the body mass,the holding power and the pulse amplitude of the model mice were significantly decreased on the 7 th day,10 th and 21 st day(P<0.01).No bleeding or significant changes in the spleen index was observed in the first 7 days in the model group(P>0.05).But on the 10 th and 21 st day,bleeding in different degrees were observed in the spleen and tail,the spleen index was significantly increased in the model group(P<0.01).The PLT counts of mice in the model group were significantly decreased on the 7 th,10 th and 21 st day.Conclusion:ITP mouse model with qi failing to control blood syndrome,Which was replicated by sleep deprivation combined intraperitoneal injection of anti-platelet serum,had the dynamic variational characteristic of qi deficiency-bleeding-qi deficiency-qi failing to control blood.
