摘要
[目的]研究胃癌(gastric cancer,GC)患者血清miR-551b-3p的表达,探讨其作为GC潜在诊断生物标志物的可能性.[方法]收集40例GC患者术前、40例正常健康者血清标本,通过实时定量q-PCR检测miR-551 b-3p的表达情况,绘制受试者工作曲线(receiver operating characteristic curve,ROC),确定诊断cut-off值.[结果]GC患者、正常健康者血清miR-551 b-3p表达水平分别为0.045±0.004、0.19±0.38,与正常健康者比较,GC患者的miR-551b-3p水平显著下调(P=0.001).血清miR-551b-3p的表达与GC的临床病理特征之间的相关性分析结果表明,血清miR-551b-3p表达与年龄、性别无相关性,但与肿瘤大小、侵袭深度和TNM阶段有明显相关性.使用miR-551b-3p诊断GC时,ROC下面积(area under the ROC,AUC)为0.857(95%CI:0.77~0.939,P=0.000),预测阈值cut-off值为0.0152,对应的灵敏度、特异度分别为87.5%、70.0%.[结论]血清循环miR-551b-3p可能是诊断GC的新型生物学标志物,但仍需进一步扩大样本量进临床研究验证其诊断价值.
[Objective]The aim of this study was to investigate the expression of serum miR-551b-3p in gastric cancer(GC)patients and to explore its potential as a diagnostic biomarker in GC.[Methods]The expression of miR-551b-3p was detected by quantitative reverse transcription polymerase chain reaction(q-PCR)in preoperative serum samples of 40 patients with GC and 40 healthy individuals(HC)from June 2015.to March 2019 in our hospital.The receiver operating characteristic curve(ROC)was generated,and the cutoff point of serum miR-551b-3p for the diagnosis of GC was selected.[Results]The expression of serum miR-551b-3p was significantly lower in patients with GC(0.045±0.004)than HC(0.19±0.38),P=0.001.The area under the curve(AUC)for serum miR-551 b-3p distinguishing patients with GC from HC was 0.857(95%CI:0.775-0.939,P=0.000),with a sensitivity of 87.5%and a specificity of 70%.[Conclusion]Serum miR-551b 3p may potentially serve as a diagnostic biomarker for GC.Further large sample prospective clinical research is needed to validate its role in the future.
