Over-expression of PBX3 Promotes Invasion and Migration of Esophageal Squamous Cell Carcinoma

PBX3是PBX家族成员之一,它是机体生长发育所必需的蛋白质因子。当PBX3基因表达异常时,它可导致肿瘤的发生,促进其侵袭转移。PBX3的作用在其他肿瘤研究中已有了解,但其在食管癌中的作用尚不清楚。本研究旨在通过了解PBX3在食管癌细胞中的表达及其与临床病理参数和患者愈后的关系,以及对食管癌侵袭及转移的影响。对食管癌进行PBX3的临床与细胞学研究结果显示,PBX3表达与食管癌患者浸润深度和淋巴转移有关,与患者的年龄、性别、肿瘤分期无关。PBX3表达水平越高,食管癌的恶性程度越高。进一步通过PBX3基因转染食管癌细胞株证明,该基因可促进上皮间质转化,并增强转移侵袭特性。我们的研究结果提示,PBX3与食管鳞状细胞癌的侵袭、转移及愈后有关,其可能是通过影响食管鳞状细胞癌的上皮-间质转化,促进癌细胞的侵袭及迁移。所以,PBX3有可能作为评估食管鳞状细胞癌恶性程度的指标。

PBX3 is one of the members of the PBX family,which is a factor necessary for the growth and development of the body.Aberrant PBX3 expression leads to tumor development and promotes tumor invasion and metastasis.However,the role of PBX3 in esophageal cancer remains unclear.This study aims to investigate the expression of PBX3 in esophageal cancer cells and its relationship with clinicopathological parameters and patient prognosis,as well as its effect on esophageal cancer invasion and metastasis.Clinical and cytological studies of PBX3 were performed on esophageal cancer.The results showed that PBX3 expression was related to the depth of invasion and lymphatic metastasis in patients with esophageal cancer,while had nothing to do with the patient’s age,gender,and tumor stage.The PBX3 gene was further transfected into esophageal cancer cell lines to prove that the gene enhances metastasis.We found that the higher the expression level of PBX3,the higher the malignant degree of esophageal cancer.In summary,our experimental results suggest that PBX3 is related to the invasion,metastasis,and prognosis of esophageal squamous cell carcinoma.And the underlying mechanism could be by epithelial-mesenchymal transition that promoted invasion and metastasis in esophageal squamous cell carcinoma.So PBX3 may be used as a target to evaluate the malignancy of esophageal squamous cell carcinoma.
