龙芪强肌饮对脊髓损伤大鼠腓肠肌功能及肌细胞凋亡的影响

摘要 目的探讨龙芪强肌饮对脊髓损伤大鼠腓肠肌功能及肌细胞凋亡的影响。方法将135只健康SPF大鼠随机分为空白组、模型组和龙芪强肌饮组,模型组和龙芪强肌饮组采用改良Allen’s法建立脊髓损伤模型。龙芪强肌饮组术后第1天开始给予龙芪强肌饮灌胃,持续42 d。分别于术前及术后1 d、14 d、28 d、42 d时各组随机选择15只大鼠采用BBB法进行后肢功能评分和腓肠肌肌电位测定,术后14 d、28 d、42 d完成后肢运动功能和腓肠肌肌电位测定后处死各组大鼠,取腓肠肌标本,测腓肠肌湿重,采用HE染色后测定肌细胞直径及截面积,液氮冰冻后采用TUNEL法检测肌细胞凋亡指数。结果术后1 d、14 d、28 d、42 d时,模型组和龙芪强肌饮组BBB评分和腓肠肌肌电位峰值均显著低于同期空白组(P均<0.05),但术后14 d、28 d、42 d时龙芪强肌饮组BBB评分和腓肠肌肌电位峰值均显著高于同期模型组(P均<0.05)。术后14 d、28 d、42 d时,模型组和龙芪强肌饮组大鼠腓肠肌湿重和肌细胞横截面积均低于同期空白组(P均<0.05),但龙芪强肌饮组均明显高于同期模型组(P均<0.05);模型组和和龙芪强肌饮组大鼠腓肠肌细胞凋亡指数均明显高于空白组(P均<0.05),但龙芪强肌饮组均明显低于同期模型组(P均<0.05)。结论龙芪强肌饮可改善脊髓损伤大鼠下肢功能、肌电位峰值及腓肠肌细胞萎缩,减少腓肠肌细胞凋亡可能是其重要作用机制。 Objective It is to investigate the effect of Longqiqiangjiyin decoction on gastrocnemius function and myocyte apoptosis in rats with spinal cord injury.Methods 135 healthy SPF rats were randomly divided into blank group,model group and Longqiqiangjiyin group.The rats in the model group and Longqiqiangjiyin group were used to establish spinal cord injury models by the modified Allen’s method.Longqiqiangjiyin group was treated with intragastric administration of Longqiqiangjiyin for 42 days from the first day after operation.15 rats were randomly selected in each group before operation and at 1 d,14 d,28 d and 42 d after operation,and their hind limb function score and gastrocnemius muscle potential were measured by BBB method.After the completion of detection of hindlimb motor function and gastrocnemius muscle potential at 14 d,28 d and 42 d after surgery,the rats of each group were sacrificed and their gastrocnemius muscle samples were taken to measure the wet weight of gastrocnemius muscle.The diameter and cross-sectional area ofmuscle cells were measured by HE staining.After freezing with liquid nitrogen,the apoptosis index of muscle cells was detected by TUNEL method.Results At 1 d,14 d,28 d,and 42 d after operation,the BBB scores and gastrocnemius muscle potential peaks of the model group and Longqiqiangjiyin group were significantly lower than those of the blank group at the same period(all P<0.05),but at 14 d,28 d,and 42 d after operation,the BBB score and gastrocnemius muscle potential peak of the Shilongqiqiangjiyin group were significantly higher than those of the model group at the same period(all P<0.05).At 14,28,and 42 days after operation,the wet weight and cross-sectional area of gastrocnemius muscle in the model group and the Longqiqiangjiyin group were lower than those in the blank group(P<0.05),but the Longqiqiangjiyin group was significantly higher than the model group at the same period(all P<0.05);the gastrocnemius cell apoptosis index in the model group and Heqiqiangjiyin group was significantly higher than that in the blank group(all P<0.05),but the Longqiqiangjiyin group were significantly lower than the model group at the same period(all P<0.05).Conclusion Longqiqiangjiyin can improve lower limb function,peak myoelectric potential and gastrocnemius muscle atrophy in rats with spinal cord injury,reducing gastrocnemius muscle cell apoptosis may be its important mechanism.
