滋肾骨康方联合阿仑膦酸钠维D3对骨质疏松大鼠骨密度、骨代谢的影响 被引量:2

Effect of Zishen Gukang Decoction Combined with Alendronate Sodium Vitamin D3 on Bone Mineral Density and Bone Metabolism of Osteoporotic Rats

摘要 目的探讨滋肾骨康方联合阿仑膦酸钠维D3对骨质疏松老鼠骨密度、骨代谢的影响。方法选择SD系雌性大鼠120只,全部制作绝经后骨质疏松症的病理模型,将全部的实验大鼠按照随机数字的方法分为4组,空白对照组(A组),阿仑膦酸钠维D3治疗组(B组),滋肾骨康方治疗组(C组)和阿仑膦酸钠维D3联合滋肾骨康方治疗组(D组),每组30只。A组大鼠每日灌胃冷开水,B组大鼠每日灌胃阿仑膦酸钠维D3的水溶液,C组大鼠每日灌胃滋肾骨康方的溶液,D组大鼠每日灌胃阿仑膦酸钠维D3和滋肾骨康方的溶液,连续12周。对比4组大鼠治疗前后的股骨,胫骨和第2~4腰椎的骨密度。对比4组大鼠治疗前后的血清钙,磷,碱性磷酸酶的浓度。对比4组大鼠治疗前后的尿液羟脯氨酸浓度。结果治疗后,A组大鼠的股骨,胫骨和第2~4腰椎的骨密度均降低,B组,C组,D组大鼠的股骨,胫骨和第2~4腰椎的骨密度均增高(P<0.05),在治疗后的组间比较中,数据差异有统计学意义(F=79.632,49.663,50.657,P<0.05),q检验结果显示,股骨,胫骨和第2~4腰椎的骨密度数值均为D组高于B组与C组,B组与C组高于A组(P<0.05),治疗前后,4组大鼠的血清钙,磷浓度未出现明显变化(F=1.458,2.089,P<0.05)。经过治疗后,A组大鼠的碱性磷酸酶增高,B组、C组、D组大鼠的碱性磷酸酶均降低(P<0.05),在治疗后的组间比较中,数据差异有统计学意义(F=67.571,P<0.05),q检验结果显示,碱性磷酸酶数值为D组低于B组与C组,B组与C组低于A组(P<0.05),经过治疗后,A组大鼠的尿液羟脯氨酸浓度增高,B组、C组、D组大鼠的尿液羟脯氨酸浓度均降低(P<0.05),在治疗后的组间比较中,数据差异有统计学意义(F=87.005,P<0.05),q检验结果显示,尿液羟脯氨酸浓度数值为D组低于B组与C组,B组与C组低于A组(P<0.05)。结论使用滋肾骨康方联合阿仑膦酸钠维D3治疗骨质疏松的大鼠模型,可以增加骨密度,影响骨代谢,有效防止骨质疏松的出现,但是在人体的结果尚需要深入研究。 Objective To explore the effect of Zishen Gukang Decoction combined with alendronate sodium vitamin D3 on bone mineral density and bone metabolism in osteoporotic rats.Methods A total of 120 SD female rats were selected and all pathological models of postmenopausal osteoporosis were made.All experimental rats were divided into four groups according to a random number method:the blank control group(group A),the alendronate sodium vitamin D3 treatment group(group B),the Zishen Gukang Decoction treatment group(group C)and the alendronate sodium vitamin D3 combined with Zishen Gukang Decoction treatment group(Group D),30 per group.Group A rats were intragastrically administrated with cold boiled water daily.Group B rats were intragastrically administered with alendronate sodium vitamin D3 daily.Group C rats were daily given Zishen Gukang Decoction.Goup D rats were given alendronate sodium vitamin D3 and Zishen Gukang Decoction daily.The treatment was lasted for 12 weeks.The bone mineral densities of the femur,tibia and the 2 nd to 4 th lumbar vertebrae before and after treatment in the four groups of rats were compared.The concentrations of serum calcium,phosphorus and alkaline phosphatase were compared before and after treatment in the four groups.The urine hydroxyproline concentrations before and after treatment in four groups were compared.Results After treatment,the bone mineral densities of the femur,tibia and the 2 nd to 4 th lumbar vertebrae of the rats in group A decreased.The bone mineral densities of the femur,tibia and 2 nd to 4 th lumbar vertebrae of group B,C and D increased(P<0.05).In the comparison between groups after treatment,the data was statistically significant(F=79.632,49.663,50.657,P<0.05).The results of q test showed that the bone mineral densities of femur,tibia and the 2 nd to 4 th lumbar vertebrae in group D were higher than those in group B and C,and those in group B and C were than those in group A(P<0.05).Before and after treatment,the serum calcium and phosphorus concentrations of the four groups did not change significantly(F=1.458,2.089,P<0.05).After treatment,the alkaline phosphatase of group A rats increased,and that of group B,C and D decreased(P<0.05).In the comparison between groups after treatment,the data was statistically significant(F=67.571,P<0.05).The results of q test showed that the alkaline phosphatase value was in group D lower than that in group B and C,and that in group B and C were lower than that in group A(P<0.05).After treatment,the concentration of urinary hydroxyproline in group A rats increased,and that in group B,C and D decreased(P<0.05).In the comparison between groups after treatment,the data was statistically significant(F=87.005,P<0.05).The results of q test showed that the urinary hydroxyproline concentration in group D was lower than that in group B and C,and that in group B and C was lower than that of group A(P<0.05).Conclusion The rat model of osteoporosis treated with Zishen Gukang Decoction combined with alendronate sodium vitamin D3 can increase bone density,affect bone metabolism,and effectively prevent the occurrence of osteoporosis.However,the results in humans need to be further studied.
