目的检测子宫内膜癌(EC)患者肿瘤组织中miR-410表达水平,分析其与临床病理特征及预后的关系。方法选取2011年6月至2013年6月确诊并进行手术的EC患者62例为EC组,选取同期因子宫内膜增生行手术的患者60例为对照组,利用实时定量PCR(qRT-PCR)检测子宫内膜组织中miR-410表达水平,采用Western blot法检测组织中MMP16和TIMP2蛋白水平。结果EC患者肿瘤组织中miR-410较对照组织低表达(P<0.05);肿瘤组织中MMP16蛋白高表达,TIMP2蛋白低表达(P<0.05);肿瘤组织中miR-410表达水平与患者年龄无关(P>0.05),与病理分期、分化程度、淋巴结转移、肌层浸润程度有关(P<0.05);miR-410高表达患者5年总生存率为93.33%,显著高于miR-410低表达者68.75%(P<0.05);多因素分析表明,高手术分期、淋巴结转移、高肌层浸润程度、miR-410低表达是影响EC患者不良预后的独立危险因素(P<0.05)。结论EC肿瘤组织中miR-410低表达,其表达水平与患者病理分期、分化程度、淋巴结转移、肌层浸润程度有关,是影响患者不良预后的独立危险因素之一,可能用于EC患者预后评估。

Objective To detect the expression levels of miR-410 in tumor tissues of patients with endometrial cancer(EC),and to analyze its relationship with clinicopathological characteristics and prognosis of patients.Methods Sixty-two patients with EC who were diagnosed and treated by surgery in our hospital from June 2011 to June 2013 were enrolled as EC group,and the other 60 patients with endometrial hyperplasia who underwent surgery at the same period were enrolled as control group.Real-time quantitative PCR(qRT-PCR)was used to detect the expression levels of miR-410 in endometrial tissues,and Western blot was used to detect the protein levels of MMP16 and TIMP2 in tumor tissues.Results The expression levels of miR-410 and TIMP2 protein in EC group were significantly lower than those in control group(P<0.05),however,the expression levels of MMP16 protein in EC group were significantly higher than those in control group(P<0.05).The expression levels of miR-410 in tumor tissues were not correlated with patients’age(P>0.05),however,which were related to pathological stage,differentiation degree,lymph node metastasis,and muscular infiltration degree of tumor(P<0.05).The 5-year overall survival rate of patients with high expression of miR-410 was 93.33%,which was significantly higher than that(68.75%)of pateints with low expression of miR-410(P<0.05).Multivariate analysis showed that high surgical staging,lymph node metastasis,high degree of muscular infiltration,and low expression of miR-410 were independent risk factors for EC patients with poor prognosis(P<0.05).Conclusion The low expression of miR-410 in tumor tissues of patients with EC is related to the pathological stage,degree of differentiation,lymph node metastasis,and degree of myometrial invasion.It is one of the independent risk factors for EC patients with poor prognosis,which may be used in the prognosis evaluation of patients with EC.
