Expression and clinical significance of lncRNA AFAP1-AS in endometrial carcinoma

目的研究长链非编码RNA肌动蛋白纤维相关蛋白1-反义RNA(lncRNA AFAP1-AS)在子宫内膜癌中的表达并分析其临床意义。方法收集2011年2月至2013年10月子宫内膜癌患者112例(观察组),在子宫切除术中采集子宫内膜癌变组织样品,另因阴道异常出血而进行宫腔镜刮宫活检或因子宫肌瘤需要摘除子宫的同期患者50例(对照组),术中收集正常子宫内膜组织样品,采用实时荧光定量PCR(qRT-PCR)法检测lncRNA AFAP1-AS表达情况,分析lncRNA AFAP1-AS表达水平与子宫内膜癌患者临床病理特征及预后的关系。结果观察组lncRNA AFAP1-AS在子宫内膜组织中的表达明显高于对照组(P<0.05);lncRNA AFAP1-AS在子宫内膜癌患者子宫内膜组织中表达与患者的年龄、淋巴结转移、合并高血压以及合并糖尿病无关(P>0.05),与FIGO分期、病理分级、肌层浸润有关(P<0.05);lncRNA AFAP1-AS高表达组5年存活率为41.94%,显著低于低表达组存活率(71.67%),差异有统计学意义(χ2=10.41,P<0.05);多因素分析显示,lncRNA AFAP1-AS表达和手术分期是影响子宫内膜癌患者预后的独立危险因素(HR=2.569,95%CI为1.727~3.812;HR=2.628,95%CI为1.386~4.983,P均<0.05)。结论lncRNA AFAP1-AS与子宫内膜癌的发生发展以及预后密切相关,可以作为临床诊断及预后判断的重要参考指标。

Objective To investigate the expression and clinical significance of long non-coding RNA actin fibrin-associated protein 1-antisense RNA(lncRNA AFAP1-AS)in endometrial carcinoma.Methods A total of 112 patients with endometrial cancer who were treated in our hospital from February 2011 to October 2013 were enrolled as observation group,whose endometrial cancerous tissue specimens were collected during hysterectomy,and the other 50 patients who underwent hysteroscopic curettage biopsy or hysteromyoma removal due to abnormal vaginal bleeding were enrolled as control group,whose normal endometrial tissue specimens were collected during operation.The expression levels of lncRNA AFAP1-AS were detected by real-time fluorescence quantitative PCR(qRT-PCR),moreover,the correlation between the expression of lncRNA AFAP1-AS and the clinicopathological characteristics and prognosis of patients with endometrial cancer was analyzed.Results The expression levels of lncRNA AFAP1-AS in observation group were significantly higher than those in control group(P<0.05).The expression levels of lncRNA AFAP1-AS in endometrial tissues of patients with endometrial cancer were not related to patient’s age,lymph node metastasis,complicated hypertension and complicated diabetes mellitus(P>0.05),however,which were related to FIGO stage,pathological grade and myometrial invasion(P<0.05).The 5-year survival rate in patients with high expression of lncRNA AFAP1-AS was 41.94%,which was significantly lower than that(71.67%)of patients with low expression of lncRNA AFAP1-AS(P<0.05).The multivariate analysis showed that the expression levels of lncRNA AFAP1-AS and surgical stage were independent risk factors for the prognosis of patients with endometrial cancer(P<0.05).Conclusion LncRNA AFAP1-AS is closely related to the occurrence,development and prognosis of endometrial cancer,which can be regarded as an important reference index for clinical diagnosis and prognosis judgment.
