摘要
为了对牛种布鲁菌2308株基因组中的sRNA进行预测和鉴定,提取牛种布鲁菌2308株的总RNA,进行RNA-Seq高通量测序,基于测序结果采用sRNA-Detect算法进行sRNA的预测,并采用反转录PCR对sRNA进行验证。结果显示,共预测筛选出3523条候选的sRNA,采用反转录PCR验证出14个sRNA可以表达,其中13个sRNA的正常表达与Hfq伴侣蛋白密切相关。本研究在牛种布鲁菌中预测出3523条sRNA,Hfq蛋白可以调控部分sRNA的正常表达。
To identify sRNA in the genome of B.abortus strain,the total RNA of B.abortus 2308 were extracted and transcriptome samples were prepared.The candidate sRNA were predicted based on the results of RNA-seq and sRNA-Detect algorithm.To verify the accuracy of prediction result,14 candidate sRNA were selected randomly,and verified by RT-PCR.At last,the relationship between Hfq chaperone protein and these sRNA was also studied.The results showed that 3523 candidate sRNA were screened and 14 of them were verified by RT-PCR.In addition,the expression of 13 out of 14 sRNA was significantly reduced in the hfq mutant strain.In total,our study indicated that 3523 candidate sRNA were identified in Brucella abortus 2308.And the Hfq chaperone protein played important roles in normal expression of various sRNA.
