广东省候鸟副溶血性弧菌的分离与鉴定

Isolation and identification of Vibrio parahaemolyticus from migratory birds in Guangdong Province,China

摘要 为了解广东省候鸟副溶血性弧菌及其携带的毒力基因、抗生素敏感性和遗传进化特征,收集广东省候鸟粪便,用国标法和toxR基因PCR检测的方法进行分离鉴定获得副溶血性弧菌;通过PCR对毒力基因进行鉴定、用Etset测定MIC值,通过ERIC-PCR对副溶血性弧菌进行遗传进化分析。结果共获得122株副溶血性弧菌,这些细菌仅含有tlh(不耐热溶血素基因),而tdh(耐热溶血素基因)与trh(耐热溶血素相关毒素基因)等均为阴性。通过ERIC分群发现福田红树林保护区与湛江的鹭源菌株和湛江雷州的鹭源菌株具有高度相似性,深圳华侨城湿地菌株分布在3群,与雷州的鹭源相似度较高,为95%;福田红树林保护区分离株与深圳华侨城湿地菌株相似度大于75%。广东省候鸟携带的副溶血性弧菌具有较为丰富的遗传多样性,毒力基因携带单一,保持着较高的药物敏感性。 To understand the situation of Vibrio parahaemolyticus carried by migratory birds in Guangdong Province,identify the virulence genes and drug susceptibility of Vibrio parahaemolyticus carried by them,and analyze the genetic evolution of migratory bird Vibrio parahaemolyticus source by ERIC-PCR method combined with the water environment in which migratory birds live.Vibrio parahaemolyticus was isolated from the fecal samples of migratory birds in Guangdong Province.Identify virulence gene and use the Etset strip to determine the MIC value to define drug resistance and genetic grouping by ERIC-PCR.122 strains of Vibrio parahaemolyticus were isolated and identified,only contained the tlh gene,both the tdh and the trh were negative.Discovered by ERIC,isolates from the water samples in the Futian Mangrove Reserve are highly similar to the strains isolated from heron in Zhanjiang and Leizhou.The strains isolated from the Shenzhen Overseas Chinese Town(OCT)wetlands are distributed in the same group with the heron source in Leizhou with a similarity of 95%.The strains isolated from the water samples of the Futian Mangrove Reserve have a similarity to the strains isolated from the water samples of Shenzhen OCT.The genetics of Vibrio parahaemolyticus in Guangdong Province is rich in genetic diversity and evolves rapidly.It may be through the spread of water samples by herons and then spread by wild ducks and crickets.
