胃癌中RNA甲基化酶NSUN2的表达及其临床意义

Expression of RNA methylase NSUN2 in gastric cancer and its clinical significance

目的探讨胃癌中RNA甲基化酶NSUN2的表达及其临床意义。方法收集109例胃癌采用免疫组化SP法检测癌组织及对应癌旁组织中NSUN2的表达,并分析NSUN2表达与临床病理特征及预后生存时间的关系。利用qRT-PCR及Western blot法检测20对新鲜冷冻胃癌组织及对应癌旁组织中NSUN2的表达。结果NSUN2在胃癌组织的高表达率为65.1%(71/109),在癌旁组织中的高表达率为15.6%(17/109),两组差异有统计学意义(P<0.05)。qRT-PCR和Western blot证实NSUN2 mRNA在胃癌组织中高表达。胃癌中NUSN2高表达与淋巴结转移、浸润深度、远端转移、脉管浸润、神经侵犯、临床分级、TNM分期及患者预后生存时间明显相关(P<0.05);与患者年龄、性别、肿瘤位置和直径无关(P<0.05)。结论NSUN2表达与胃癌TNM分期和侵袭性显著相关,NSUN2表达影响患者预后,NSUN2对于胃癌诊断和预后的评判具有一定的价值。

Purpose To explore the expression and clinical significance of RNA methylase NSUN2 in gastric cancer(GC).Methods 109 cases of GC were collected,immunohistochemical method was used to detect the expression of NSUN2 in tissues and corresponding tissues adjacent to cancer,and the relationship between NSUN2 expression and clinicopathological characteristics of GC and prognosis was analyzed.The expression of NSUN2 in 20 pairs of fresh frozen GC tissues and corresponding adjacent tissues was detected by qRT-PCR and Western blot.Results The high expression rate of NSUN2 in GC tissues was 65.1%(71/109)and 15.6%(17/109)in adjacent tissues,the difference between the two groups was statistically significant(P<0.05),confirmed by qRT-PCR and Western blot that showed that NSUN2 mRNA was highly expressed in GC tissues.The high expression of NUSN2 was significantly correlated with patients lymph node metastasis,depth of invasion,distal metastasis,vascular invasion,nerve invasion,clinical grade,TNM stage,and prognosis survival time of patients(P<0.05),and not related to age,gender,tumor location and the diameter of the tumor(P<0.05).Conclusion NSUN2 expression is significantly associated with TNM stage and invasiveness of GC.NSUN2 expression affects the prognosis of patients.NSUN2 can be used to evaluate the condition and prognosis of GC patients.
