甜荞多糖提取工艺的优化及其颗粒剂的制备 被引量:3

摘要 目的:优选甜荞多糖水提醇沉的最佳提取工艺及其颗粒剂的最佳制备工艺,期望为甜荞多糖的开发与应用提供实验依据。方法:首先利用紫外分光光度法结合均匀设计U5(53),用苯酚-硫酸法测定甜荞中多糖含量为指标,考察料液比、提取时间和提取温度对甜荞中多糖水提醇沉的影响;再以可溶性淀粉、糊精为稀释剂和赋形剂,70%乙醇为润滑剂制粒,并测定其溶解性、成型性和稀释性,综合评定多糖颗粒的稳定性。结果:最佳提取工艺为10倍量水在80℃下提取120 min,该条件下提取多糖含量实测值为12.27 mg/g;最佳制粒工艺荞麦多糖、可溶性淀粉和糊精用量比例为2∶1∶5,并使用70%乙醇作为润湿剂制得颗粒剂成型性好、流动性好且不易粘连,溶化速度快且静置后无沉淀。结论:优选出的甜荞中多糖的最佳提取工艺可靠,为其后期的研发奠定基础。 Objective:The extraction process of polysaccharide bucwheat was optimized,and provide experimental basis for the development and application of Fagopyrum esculentum moench.Methods:Content of polysaccharide buckwheat was determined by phenol-sulfuric acid method,and the effects of ratio of material to liquid,extraction time and extraction temperature were investigated.Then following methods were used for granulation;the proportion of buckwheat polysaccharide,soluble starch and dextrin was 2∶1∶5,and 70%ethanol was as lubricant.Finally,solubility,formability and dilution of granules were measured,and the stability was evaluated comprehensively.Results:The optimum extraction process was 20 times of water at 80℃for 120 min.The granules have good formability,fluidity and solubility.Conclusion:In this experiment,the optimum extraction process of polysaccharide bucwheat from Fagopyrum esculentum moench is reliable,and the preparation method of granules is stable,which lay a foundation for later research and development.
