枸杞多糖对大鼠任意皮瓣术后炎性反应的影响 被引量:2

摘要 目的观察枸杞多糖对大鼠任意皮瓣术后炎性反应的影响,探讨其可能的机制。方法将40只雄性SD大鼠随机平均分为5组,A组未行皮瓣手术的SD大鼠正常饲养,B组于造模前1 d用生理盐水连续灌胃10 d,C组、D组、E组于模型制备前1 d用LBP溶液连续灌胃10 d,剂量分别为0.2、0.4、0.8 g/(kg·d)。在大鼠背部设计4 cm×2 cm的任意皮瓣并原位缝合,记录各组皮瓣的一般情况,测算皮瓣成活率;HE染色后镜下观察皮瓣炎性细胞浸润及血管新生情况,用ELISA法检测各组大鼠血清中TNF-α、IL-6、MIP-1α的含量。结果与A相比,造模后的各组皮瓣均有不同程度的红肿,并逐渐减轻,7 d后各实验组大鼠皮瓣呈现不同程度的坏死;B、C、D、E组的皮瓣成活率分别为15.92%、47.52%、64.14%、83.68%,组间比较差异有统计学意义(P<0.05)。各实验组大鼠皮瓣组织切片可见不同程度炎性细胞浸润,出现新生毛细血管;各实验组造模后12、24、48、72 h及7、10 d检测炎性因子的水平较A组高,差异有统计学意义(P<0.05),实验组组间比较差异有统计学意义(P<0.05)。结论大鼠行任意皮瓣修复术后炎性反应加重,枸杞多糖可以下调皮瓣术后炎性因子的表达,减轻缺血-再灌注损伤(IRI),促进皮瓣成活。 Objective To observe the effects of Lycium Barbarum Polysaccharides(LBP)on the inflammatory in rats with random flap after operation and explore the possible mechanism.Methods 40 male SD rats were randomly divided into 5 groups.Rats in Group A unsuffered flap surgery were fed normally.Rats in Group B were gavaged with normal saline for ten days before modeling.Rats in groups(C,D,E)were gavaged with different doses of LBP solution[0.2 g/(kg·d),0.4 g/(kg·d),0.8 g/(kg·d)].A free flap(4cm×2cm)was designed on the back of the rats and sutured in situ.The general condition of the flaps in each group were recorded and the survival rate of the flaps were calculated.The inflammatory cell infiltration and angiogenesis of the flaps were observed under the microscope after HE staining,and the contents of TNF-α,IL-6 and MIP-1αin the serum of each group were detected by ELISA.Results Compared to group A,there were some different degrees of redness and swelling in all flaps of experimental groups.After 7 days,there were some different degrees of necrosis in experimental groups.The survival ratios of flap in the experimental groups(B,C,D,E)were 15.92%,47.52%,64.14%,83.68%respectively.There were statistical significances among groups(P﹤0.05).In each experimental group,there were inflammatory cells infiltration and new capillaries in different degrees.The levels of TNF-α,IL-6,MIP-1αin the experimental groups were higher than that in group A.There were statistical significances(P﹤0.05).Conclusion The inflammatory response of rats after flap repair is more serious.LBP can down-regulate the expression of inflammatory factors,weaken the effect of ischemia-reperfusion injury(IRI)and promote the survival of flap.
