摘要
采用超声波法提取圆齿野鸦椿果皮总多酚(TPEP),通过Box-behnken法对提取工艺进行优化,获得TPEP提取工艺:乙醇浓度45%,液料比31∶1,提取温度50℃,提取时间70 min,超声功率300 W.此条件下TPEP提取率高达(8.28±0.14)%.以脂多糖诱导小鼠巨噬细胞RAW264.7构建体外炎症细胞模型,测定TPEP作用下细胞上清液中IL-6、PGE2、TNF-α、NO的分泌量,发现TPEP可显著抑制这些炎性因子的表达,体现了良好的抗炎能力.
Ultrasound method was adopted to extract the total polyphenols from Euscaphis konishii Hayata pericarp(TPEP).Based on Box-behnken experiment,the optimal extraction process of TPEP was to use 45%ethanol solution at liquid-material ratio of 31∶1,and extract under 50℃at ultrasound power of 300 W for 70 min,reaching a extraction rate of(8.28±0.14)%.Meanwhile,lipopolysaccharide(LPS)-induced mice macrophage RAW264.7 cell model was established to determine the secretion of cellular inflammatory factors in the supernatant of TPEP.The results showed that TPEP can greatly reduce the secretion of cellular inflammatory factors,including IL-6,PGE2,TNF-αand NO,indicating a favorable anti-inflammatory ability.
