南非叶总黄酮的体外抗肿瘤活性研究 被引量:4

摘要 目的以食管癌细胞为主,全方位的探究南非叶总黄酮的体外抗肿瘤作用。方法以人胃癌细胞、人食管癌细胞、人肝癌细胞以及人乳腺癌细胞为对象,采用CCK8法检测南非叶总黄酮对上述细胞增殖抑制活性,观察细胞所呈现出的形态,采用CCK8法评价南非叶总黄酮对食管癌细胞(ECA109)的影响,并采用流式细胞法检测细胞凋亡情况。结果南非叶总黄酮可以显著的抑制人食管癌ECA109细胞增殖,同时还呈现出量效以及时效关系,当南非叶总黄酮浓度为500μg·mL-1,持续72h作用,抑制率极限情况下可以达到86.9%;通过荧光倒置显微镜进行观察后可以发现,细胞有着相对更少的数量,细胞质表现出回缩;流式细胞法检测显示,G 0/G 1期基因组有着相对更少的水平,S期与G 2/M期基因组有着相对更高的水平。结论南非叶总黄酮在体外对食管癌ECA109细胞具有较强的抑制作用,可诱导细胞凋亡。 OBJECTIVE To study the anti-tumor activity of total flavonoids of Bitter Leaf on esophageal cancer cell(ECA109).METHODS CCK8 was used to detect the proliferation inhibitory activity of Bitter leaf total flavonoids on four kinds of cells such as human gastric cancer cell line(BGC-823),human esophageal cancer cell line(ECA109),human liver cancer cell line(HepG-2)and human breast cancer cell line(MDA-MB-231).The effect of Bitter leaf total flavonoids on ECA109 was evaluated by cell morphology observation and CCK8.And apoptosis of tumor cells was detected by flow cytometry.RESULTS The inhibition activity of total flavonoids on ECA109 cells was the highest,with a certain dose-dependent relationship and aging-dependent relationship.When the total flavonoids concentration was 500μg·mL-1,the highest inhibitory rate reached 86.9%in 72h.The results showed that the number of cells decreased and the cytoplasm retraction was observed under the inverted fluorescent microscope.The results of flow cytometry showed that the G 0/G 1 genome level decreased and the genome level of S and G 2/M cells increased.CONCLUSION Total flavonoids of Bitter Leaf have a strong inhibitory effect on ECA109 in vitro,which can induce apoptosis of tumor cells.
