摘要
目的建立高效液相色谱-示差折光检测器(HPLC-RID)法测定复方金刚烷胺氨基比林片中盐酸金刚烷胺的含量。方法采用Eclipse Plus C 18色谱柱(4.6mm×250mm,5μm),流动相为甲醇-水(10∶90),柱温30℃,进样样品的体积为20μL,流速1.0mL·min^-1,检测器为示差折光检测器,检测池温度30℃。结果盐酸金刚烷胺的线性范围是2.5012~12.506mg·mL^-1(r=0.9999),平均回收率为100.12%,RSD为0.2%(n=6)。结论该方法不仅简单、方便、快捷,而且结果准确、重现性好,能够用来测定盐酸金刚烷胺的含量。
OBJECTIVE Determination of amantadine hydrochloride in Compound Amantadine Hydrochloride Tablets by HPLC with differential refractive index detector.METHODS Eclipse Plus C 18(4.6mm×150mm,5μm)column was used with methanol and water(10∶90)asthe mobile phase,the column temperature was 30℃,the volume of sample sample was 20μL,the flow rate was 1.0mL·min^-1,the detector was RID,and the detection pool temperature was 30℃.RESULTS Amantadine hydrochloride presented a good linear correlation in the range of 2.5012-12.506mg·mL^-1(r=0.9999).The recovery was 100.12%,with RSD of 0.11%(n=9).CONCLUSION The method is simple,convenient,quick,accurate and reproducible,and can be used for the determination of amantadine hydrochloride.
